Description
Provide complete sequence of aflatoxin biosynthetic gene cluster from 5 isolates of Apsedrgillus with an accuracy better than 1 error per 2000 pb (99.95%). The sequence is between 65 and 75 KB. ARS will provide high quality genomic DNA sequence information derived from a single aflatoxin producing strain. Sequence information is available for all genes in the cluster, as well as, most non-coding regions. Sequence is available for both ends and the middle of the cluster. The contractor will develop the necessary cloning strategy to separate the required sequence from other sequences, carry out the sequencing with sufficient coverage to provide the requisite accuracy, and perform sequence assembly. Finally, the contractor will provide the sequence in the format accessible to common databases, such as Genbank. Contractor will provide: 1) Cosmid, fosmid or BAC genomic libraries for each isolate. 2) Isolate clones from these libraries containing the entire 75 kb aflatoxin biosynthetic cluster using known sequence information for comparison. 3) Shotgun subclone form the socmid clones. 4) Sequence subclones to at least 6X coverage to provide sequence accuracy of >99.95%. 5) Assemble sequences from subclones. 6) Provide sequence in database accessible format. We would prefer a strategy based on cloning and shotgun sequencing. However, other strategies (i.e. long range PCR with high fidelity polymerase) will be considered.
