Description
The USDA, Agricultural Research Service in Albany, Ca. has a requirement for a benchtop mass spectrometry System. The LCQ Deca ion-trap mass spectrometer with standard electospray source, nano spray source, Xcalibur software and Turbo SeQuest software, Surveyor pump, Surveyor autosampler & Surveyor detector meets our needs The characteristics of the system include a liquid chromatograph mass spectrometer LC/MS system capable of analyzing complex mixtures of proteins, both as intact proteins and as proteolytic digest mixtures of proteins introduced as liquid solutions. Mass spectral analysis to be carried by electrospray ionization of samples and accommodating flow rates in the nano-liter per minute range and above. An autosampler, fully integrated with the mass spectrometer data system, for unattended introduction of samples either by flow injection or in conjunction with a liquid chromatograph. A complete data system for control of the mass spectrometer and the associated liquid chromatograph. The capability of carrying out MS n, where n is at least 3, i.e. MS/MS/MS for the purpose of providing de-novo sequence information on unknown peptides obtained as digest of whole proteins. A protein identification computer system capable of protein database query, and using postulates for amino acid sequences obtained from analysis of MS/MS spectra multiply charged ions obtained by electrospray ionization. A suite of deconvolution software suitable for assignment of molecular weight values to intact proteins in the 10000 to 100000 dalton ranges ionized using electrospray ionization. An LC interface suitable for use with nanoflow HPLC, and including a video observation system to aid facilitate the alignment of the electrospray needle tips. An orthogonal introduction system for delivering ions produced by electrospray into the mass analyzing section of the mass spectrometer, so as to obtain sensitivity in the 100 femptomole/microliter range or lower for peptides up to 2000 daltons molecular weight for MS/MS spectra. Demonstrated sensitivity for reserpine, a universal standard for MS/MS analysis, of at least 50 pg/microliter, using electrospray MS/MS analysis of its molecular ion at a flow rate of 400 microliters/minute. Spectra obtained in this manner to include peaks at m/z 397 or 408 of at least 100:1 S/N ratio. The mass spectrometer will be used to investigate the protein composition of bacterial and viral pathogens.
