- This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. Offers are due in the designated contracting office by 11:00 a.m. local time, January 13, 2003. Offers may be FAXED, E-Mailed, Express Mailed, Mailed, or hand delivered. The solicitation number is 263-03-P-(SC)-0037, and is issued as a Request for Proposal (RFP). The solicitation/contract will include all applicable provisions and clauses in effect through FAC 10 effective 12/23/2002. The North American Industry Classification System (NAICS) code is 541710 and the size standard is 500 employees. This solicitation is a small business set-aside. NIH has a need for an Expression Analysis of RNA samples from Mouse inner ear. The analysis shall include, but is not limited to the following: 1. The Megaclone Library construction shall consist of the following: a. Generation of Megaclone library from inner ear RNA. b. Titering of library and DNA isolation. c. PCR amplification. d. Preparation of cDNA for bead loading. e. Bead loading by hybridization. f. Collection of the loaded beads. g. Post sort enzymatic processes. 2. The massive parallel signature sequence (MPSS) Sequencing of 1 million signature tags shall be as follows: a. Loading of the flow cells. b. Enzymatic preparation of the loaded beads. c. Hybridization of the encoadopters to detect first four bases. d. Image process. e. Repeat cycle up to 17 bases. f. Base calling and clustering. g. Run group formation from multiple flow cells. h. Data analysis and annotation. 3. Total RNA from 200 mouse organs of Corti from each of the three stages (P0, P7 and either E18 or P16 B6) will be supplied to the contractor. Dissections of organs of Corti from these three stages will be completed and total RNA isolated in the Laboratory of Molecular Genetics (LMG) by Drs. Belyantseva, Battey, Friedman and Morell. Total RNA samples will be supplied, one at a time, to the contractor in mid January, mid February and mid March 2003. 4. The specific aims of this project are to deeply probe cDNA libraries from the mouse organ of Corti in order to: a. Identify novel genes expressed in the organ of corti. b. Quantify the levels of virtually all genes expressed in the organ of Corti. c. Describe qualitatively the genes expressed differently during the late stages of development of the organ of Corti. 5. The contractor will be provided with total RNA and will isolate poly A+RNA from our samples, prepare a cDNA library, amplify the cDNA, and massive parallel signature sequence (MPSS) to obtain at least 1 million signature tags from each of the three libraries. Work shall be completed by the contractor within 8 to 12 weeks of receipt of each RNA sample from the LMG. FOB Destination is required and delivery is to Dr. Battey, NIH, BLDG 31 Room 3C02. The Contractor shall submit a firm offer for all required services including any discounts. Offers shall detail descriptions of services offered, delivery schedule, and past performance data for similar work including telephone/FAX numbers and e-mail addresses. Contractor will be selected based on offered services, past performance data, and price. A completed copy of FAR provision 52.212-3, Offer representations and Certifications ? Commercial Items, should be submitted with the proposal.
