- The National Cancer Institute (NCI), Division of Cancer Epidemiology and Genetics (DCEG), Radiation Epidemiology Branch (REB) intends on entering into a sole source contract with Harvard School of Public Health, Environmental Epidemiology, 665 Huntington Avenue, Boston, MA 02115. REB conducts interdisciplinary epidemiological studies to identify and quantify the role of ionizing radiation in relation of risk of cancer. An investigation of cancer risks in a large cohort of U.S. Radiologic Technologist were certified for two of more years during 1926-82, found elevated risks for non melanoma skin cancer among subjects who began working as radiologic technologist before 1950, and risk rose with increasing number of years worked before 1950. Non melanoma skin cancer (which includes the two major types -basal cell skin cancer and squamous cell skin cancer) is the most frequently occurring malignancy, with more than one million new cases diagnosed annually in the U.S. The major known cause of non melanoma skin cancer is ultraviolet radiation from sun exposure, particularly in persons with red or blond hair, blue eyes, and/or skin that burn easily and does not tan. High levels of ionizing radiation have been associated with increased risk of non melanoma skin cancer, but risks of non melanoma skin cancer have not been extensively studied at moderate or low levels of ionizing radiation exposure. Ionizing radiation exposure results in cellular damage at the DAN level, primarily observed as single strand breaks and oxidative base damage. Prevention of neoplasia is dependent upon the proficient repair of such damage to avoid the accumulation of somatic mutations. XRCCI is a protein involved in both the base excision and strand break repair pathways, making it an attractive candidate for the investigation of ionizing radiation susceptibility. There are nine (9), published polymorphisms in the XRCCI gene that result in an amino acid substitution. Three of these (codon 194, 280 and 399) occur with a population prevalence amenable to epidemiologic study. In order for REB to evaluate risks and dose-response effects for ionizing radiation exposure, ultraviolet radiation exposure from the sun, constitutional factors (such as hair color, eye color, and skin color), and genetic factors in the etiology of non melanoma skin cancer, a series of pilot studies is required to provide important information for developing the study protocol. The services in this study will be carried out using DNA and questionnaire data form subjects participating in an existing Case-Control Study in New Hampshire. The Department of Cancer Cell Biology, at Harvard School of Public Health is the only source known to the NCI with the expertise in molecular epidemiology and genetics. Harvard School of Public Health is the only source known to NCI that has access to the samples, expertise and qualified personnel that can run the statistical aspects of the proposed requirement. The statement of capabilities and any other information must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform the requirement.
