Description
NAICS Code: 541710 To develop bioindicators for aquatic toxicity applications, the US Environmental Protection Agency intends to award a purchase order under simplified acquisition procedures to Amersham Biosciences. The associated North American Industry Classification System (NAICS) code is 541710 which has a size standard of 500 employees to qualify as a small business. Amersham Bisociences will develop a database to construct a microarray that would provide an important tool to achieve the Agency's goal of providing a tool to assess risk for water resources. Amersham has developed a 3-dimensional coating to synthesize DNA oligos onto microarray slides affording a robust, reproducible, optimal hybridization platform to ensure a dynamic range of detection and results in more useable data per bioarray. Amersham will provide custom synthesis of a fathead minnow array and provide quality control validation to guarantee accurate and reliable results. The following are requirements for this particular requirement: 1.Custom bioarrays need to have a 3-dimensional slide coating comprised of a polymer with amine-reactive groups for covalent attachment of 5' amino-labeled oligonucleotides. 2. Custom oligonucleotides need to be designed based on provided DNA sequences, and need to be screened in silico and tested under standard expression conditions to ensure optimal and accurate hybridization performance. 3. 100% post dispense inspection needs to be performed using tracer dyes for the identification and tracking of any manufacturing defects. 4. Bioarrays should be able to detect as low as a single transcript per cell with as little as 2 micrograms of total RNA starting material. 5. The dynamic range needs to be greater than 3 orders of magnitude which gives more usable data per bioarray. 6. Each bioarray should be set up to ensure uniform hybridization which reduces variability between users. 7. Array set-up and design needs to be compatible with the Affymetrix 528 custom slide analyzer. The EPA has identified Amersham Biosciences as the only organization with the qualifications and products to perform the task within the prescribed time frame, i.e., FAR 6.302-1 "Only one responsible source and no other supplies or services will satisfy Agency requirements". Interested parties who can meet the above requirements may submit qualification statements no later than 15 days after the date of this publication.
