- Description
- The National Institutes of Health (NIH), National Cancer Institute (NCI), Division of Cancer Etiology and Genetic (DCEG), Viral Epidemiology Branch (VEB) intends to procure laboratory services to perform quantitative PCR (polymerase chain reaction) testing for detection of polyomavirus DNA in human specimens. A role for viruses in the etiology of childhood cancer has been long posited. Polyomaviruses represent attractive candidates for causative agents of childhood cancer. Recently, investigators described detection of BK virus DNA in neuroblastoma tumors from children. NCI will extract DNA from specimens obtained from neuroblastoma patients, children without neuroblastoma, and laboratory controls. The University of Colorado will utilize real-time PCR to detect and quantify BK virus DNA in these DNA extracts. They will utilize primers from two regions of the polyomavirus genome, namely the T antigen and VP1 genes. They will utilize PCR assays with demonstrated sensitivity of at least 100 viral copies in 500 ng of genomic DNA. Amplified products consistent with BK virus shall be sequenced for confirmation. The laboratory shall also perform similar PCR assays for JCV, as a negative control. Finally, to assist the NCI in exploratory studies of the role of other polyomaviruses in human cancer, the PCR testing laboratory shall perform similar PCR testing for simian virus 40 (SV40) and lymphocytic polyomavirus (LPV). Thus, Dr. Garcea?s labotaroty shall perform quantitative PCR testing for DNA from two gene regions (T antigen and VP1) of these four polyomaviruses (BK, JC, SV40, and LPV). Dr. Garcea?s laboratory at the University of Colorado is the only laboratory known to the end user that can perform these tests. This laboratory developed and validated these assays. This is not a request for competitive quotation. However, if any interested party believes it can meet the above requirement, it may submit a statement of capabilities. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow the NCI researcher to determine if the party can perform this requirement. An original and one copy of the capability statements must be received in the contracting office by 1:00 p.m. EDT, on May 4, 2004. A determination by the Government not to compete this proposed requirement based upon responses to this notice is solely within the discretion of the Government.
