Description
This notice is a combined synopsis/solicitation for commercial items prepared in accordance with format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; quotes are being requested & a written solicitation will not be issued. Submit offers on RML-RFQ-4053. The solicitation documents and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2001-24 dated 6/18/04. This acquisition will be processed under Simplified Acquisition Procedures (SAP) and is not a Small Business Set-Aside. The North American Industry Classification System (NAICS) code for this procurement is 541710 and the small business size is 500. SCHEDULE: The Rocky Mountain Laboratories, (RML), Laboratory of Human Bacterial Pathogenesis (LHBP) performs basic research contributing to the development of diagnostic reagents, therapeutics and vaccines against human bacterial pathogens. LHBP has a need for protein purification, crystallization and low resolution structure analysis of a bacterial pathogen protein. This shall be a six(6) month contract, where the final product shall be the low resolution structure of one bacterial protein. LHBP requires large scale production of the bacterial protein, purification, crystallization in the presence and absence of a defined ligand, analysis by X-ray scattering, binding reaction analysis, and consultation on amino acid substitutions and ATPase assays in vitro. The contractor shall provide personnel for the work effort. The contractor shall work closely with LHBP staff members to insure that the project is progressing and that problems or issues that arise are discussed. The contractor shall keep all NIH, RML-derived data secure and shall not release, make public or maintain any of the RML data beyond this contract effort. If Internet connectivity is needed in order to perform the work, for transfer of files, or other efforts, contractor computers shall be secure and in compliance with RML-IT security procedures and policies. Internet connectivity and/or remote access for the purpose of performing the work, for transfer of files, or other efforts, shall be coordinated with NIAID IT staff to ensure security and compliance with NIAID IT procedures and policies. The Contractor shall produce/provide the following reports and deliverables. 1) Construction of Plasmids for expressing the bacterial protein that contains the receiver domain and adjacent linker element; 2) Purification of the protein by performing 20L fermentations to generate E.coli cell paste, followed by FPLC chromatography, gel filtration and/or dialysis shall be performed; 3) The contractor shall accomplish characterization of the protein by small angle X-ray scattering experiments to determine low resolution structure of the protein; 4) The Contractor shall accomplish Synthesis of 15N-labeled protein and titration of binding reactions with Mg2+ and BeF3- using HSQC NMR chemical shift data; 5) The contractor shall accomplish performance of crystallization trials of the bacterial protein in the presence of and absence of Mg2+ and BeF3-. If #5 is successful, then #6 and #7 will be attempted; 6) Following the production of diffraction crystals, collection of diffraction data and phase information by molecular replacement or MAD or SAD phasing methods; 7) Following obtainment of phase information, refinement of the high resolution model; 8) Suggestions will be provided on amino acid substitutions to the project officer; 9) An ATPase assay shall be provided to the project officer along with training on use of the assay with RML-constructed protein amino acid substitution variants. Offerors must include with their offer a completed copy of the provisions at FAR 52.212-3 Offerors Representations and Certifications—Commercial Items. The contractor shall be required to complete on-line Security Training to insure compliance with RML-IT security procedures and policies. (B)(1) By submission of an offer, the offeror acknowledges the requirement that a prospective awardee shall be registered in the CCR database prior to award, during performance, and through final payment of any contract, basic agreement, basic ordering agreement, or blanket purchasing agreement resulting from this solicitation; Award will be based on the capability of the item offered to meet the above stated salient characteristics, technical evaluation, price, and delivery.
