- Description
- The National Cancer Institute (NCI), Center for Cancer Research (CCR), plans to purchase on a sole source basis an Amersham Biosciences AKTA-FPLC system from Amersham Biosciences/GE Healthcare, P.O. Box 1327, 800 Centennial Avenue, Piscataway, NJ 08854. The supplies and services herein are being procured in accordance with FAR Part 12. The North American Industry Classification System Code is 334516 and the business size standard is 500 employees. The Dermatology Branch, CCR, NCI has broad responsibilities for studying fundamental and clinical aspects of neoplastic and inflammatory skin diseases, as well as normal skin function. We are proposing to purchase the Amersham Biosciences AKTAFPLC unit as a fully integrated instrument system used for purifying biomolecules from complex starting materials derived from cell lysates, tissue homogenates, recombinant protein expression systems and other sources. This system is required to provide the versatility for the different research needs of the branch members, ranging from antibody purification, purification of recombinant fusion proteins, and complex purifications of signaling enzymes from tumor cell lysates. The Dermatology Branch requires the Amersham Biosciences AKTA-FPLC system for purification of normal and aberrant signaling proteins from the complex mixture of proteins comprising human melanoma tumor cell lysates and other cell lysates, for the efficient purification of bacterially-generated recombinant fusion proteins, and for the affinity purification of antibodies. We require the ability to purify biomolecules from complex starting materials derived from cell lysates, tissue homogenates, recombinant protein expression systems and other sources. The system we will use must be able to perform separations based on a variety of sample characteristics, such as molecular weight, net surface charge, hydrophobicity, while retaining biological activity. We require a flow rate range of .05-20mls/min at pressures ranging from 0-700 psi.. The entire wetted flow path system must be a two-pump gradient system, with dynamic mixing, as evidence suggests that this configuration produces the highest accuracy and precision in gradient formation. The system must be inert to halides. The system pumps shall allow operation of the system at zero backpressure to ensure system compatibility with standard low pressure chromatography supports. The controller must allow programs to be time-based, volume-based, or column-volume based, and must allow user intervention of a running method (e.g., change of flow rate) without stopping or pausing the run. The system must include a fraction collector that is fully integrated into the system and fully controllable via the system control software. Amersham Biosciences/GS Healthcare is the only source known that can provide this system. This is not a request for competitive quotation. However, if any interested party believes it can perform the above requirement, it may submit a statement of capabilities. The statement of capabilities and any other furnished information must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform this requirement. Capability statements must be received in the contracting office by 1:00 PM EST, September 9, 2004.
