- This is a combined synopsis solicitation for commercial services, prepared in accordance with the format in FAR Part 12.6, as supplemented with additional information included in this notice. THIS ANNOUNCEMENT CONSITUTES THE ONLY SOLICITATION AND A SEPARATE SOLICITATION WILL NOT BE ISSUED. This solicitation includes applicable provisions and clauses in effect through FAR FAC 2001-25 (11/2004). The North American Industry Classification System Code is 621511 and the business size standard is $11.5M. The Government will furnish all necessary materials, equipment and facilities for the contractor to perform the required services. Background information: The Neuromuscular Diseases Section, National Institutes of Neurological Disorders and Stroke (NINDS), is involved in the investigation of immunological and genetic basis of a wide range of neuromuscular diseases. The investigations involve use of tools in immunology, biochemistry, proteomics, genomics and conventional genetics. Also being conducted are several clinical trials to study the efficacy and pharmcogenetics of drugs used in the treatment of neuromuscular diseases like inclusion body myositis, stiff- person syndrome, demyelinating neuropathies and post polio syndrome. One of the major focuses is the study of the role of T cells in these diseases by isolating them and characterizing structure-function relationships. Contractor shall provide an individual with expertise in T cell cloning, expansion, T cell phenotyping, lymphocyte isolation, human skeletal muscle culture, isolation of t cells from muscle biopsy samples, protein separation, immunoblotting, and molecular biology techniques. This is a non-personal service contract. Independently, and not as an agent of the Government, the contractor shall furnish the services described in the statement of work. The contractor shall meet with the Project Officer within five (5) calendar days after contract award to discuss the contract and to ensure mutual understanding of operating procedures and the services to be performed. Contract Requirements: A. Specific Tasks. 1) The contractor shall conceptualize, develop and organize experimental protocols in the study of neuromuscular diseases. B. Specific techniques and methods to be performed by the contractor. 2) Culture human skeletal muscle cells from biopsies of patients with neuromuscular diseases. The contractor should have prior experience of culturing human skeletal muscle cells from biopsies including initiating explant cultures, maintaining, splitting and various passages, 3) Cloning and culturing T cells from human skeletal muscle biopsies; isolate T cells from skeletal muscle biopsies of patients with neuromuscular diseases. 4) Clone and culture cytotoxic T cells and immunophenotype them. The contractor shall have experience in cloning and culturing cytotoxic T cells from human muscle biopsies, 5) T cell cloning from Cerebrospinal Fluid (CSF) and characterization. The contractor shall perform T cell cloning from the cerebrospinal fluid, expand and immunophenotype the cells by FACS (Fluorescence Activated Cell Sorting/Scanning), 6) Protein separation and detection techniques. The contractor shall have experience in the separation of proteins by denaturing and non-denaturing polyacrylamide gels, transferring onto appropriate membranes, and immunodetection of proteins using appropriate antibodies. This is a technique that requires excellent skill, as problems like signal to background ratios, inappropriate loading or immunodetection can ruin the experiments that use precious patient-derived biological samples. The contractor shall have experience in using human brain proteins, 7) Real time and conventional polymerase chain reaction (PCR) methodologies to quantify gene expression; isolate RNA and DNA, prepare cDNA and perform meticulous PCR experiments with large number of samples and data analysis. Very little amounts of muscle biopsies are to be used with at most, care and patience to obtain maximum yield, 8) FACS. The contractor shall analyze lymphocyte subsets, specifically T and B cells, for cell surface markers and intracellular cytokines to characterize the phenotype and function. This is accomplished by three and four color FACS analysis and immunostaining for intracellular proteins. This will also involve fractionation of subsets of T and B cells by affinity columns and magnetic separation techniques, 9) Radioimmunoassay. The contractor shall perform radioimmunoassay to quantitate GAD65 antibodies in the sera and CSF, 10) Immunoassays: The contractor shall perform ELISA (Enzyme Linked Immunosorbent Assay) experiments to quantitate cytokines and chemokines and, 11) T cell proliferation assays: The contractor shall perform thymidine uptake assays to assess T cell proliferation. Contractor shall notify the Project Officer if the assigned task was not accomplished at the end of each month. A statement regarding the appropriate reasoning shall be completed and provided to the project officer via the laboratory notebook or Microsoft word document. C. Contractor shall submit the following for each task/specific techniques and methods identified above to the NINDS Project Officer: 1) Outline of the experiments to be performed. The experimental outlines and plans are to be appropriately recorded in the assigned laboratory record book, 2) Daily documentation of the experiments conducted. The contractor shall maintain meticulous documentation of experiments performed in the assigned laboratory record book and, 3) Final report at the end of each option period. The final report may be either in electronic or in hard copy format. The report does not need to be more than one page but, shall consist of the accomplishments of the period with respect to the task assigned. Qualification Requirements: Provide documentation as it relates to the following: The contractor shall have a Master?s degree in Biomedical Science and have a minimum of 10 years experience working in a research lab. The candidate should have expertise in all the tasks outlined in the contractor requirements, A and B. In addition the following tasks/expertise the contractor shall possess: 1) Knowledgeable of scientific and medical terminology in relation to neuromuscular diseases and, 2) Expertise in the use of all of the following equipments: Perkin Elmer Micro beta, Cell harvester, MJ Research Opticon real time PCR machine, MJ Research thermal cycler, Becton Dickinson FACScaliber, film developer. Government Responsibilities: The Government shall: 1) Provide a detailed description of experiments to be performed (task), including timeline for review of drafts and expected completion schedule, 2) Review draft and final reports submitted by contractor to ensure tasks are completed correctly and on schedule, 3) Schedule face-to-face, telephone and e-mail conversations between relevant parties especially for larger, more complex projects involving collaborative efforts and to resolve unanticipated issues, if needed and, 4) Review invoices to ensure accurate billing. Period of Performance: Base Year: February 1, 2005 through December 31, 2005, Minimum number of hours 80, Maximum number of hours 2,200. Option Year 1: January 1, 2006 ? August 31, 2006, Minimum number of hours 80, Maximum number of hours 2,200. FAR Clauses 52.217-8 Option to Extend Services and 52.217-9 Option to Extend the Term of the Contract apply. Payment: The contractor shall invoice monthly for services provided. The Section Chief or an appointed person will verify the laboratory record before authorizing the payment request. Final payment will be issued after the submission and approval by the government of final document(s). BASIS FOR AWARD: Award will be made, subject to available funding and quoter responsibility, to that acceptable quote, the price or cost of which is not necessarily the lowest, but which is sufficiently more advantageous to the Government than the lowest offer so as to justify the payment of additional amounts, based on the evaluation criteria described in this section. FORMAT AND INFORMATION FOR SUBMITTING A QUOTE: Please submit ONE (1) original plus two (2) copies of entire quotation. Please submit your quotation by the following: VOLUME 1: Contract and Pricing Documents: (1) Signed SF1449 (page 1, blocks 17a, 21, 22, 23, 24, 26, 30a, 30b, 30c and other pricing information), (2) Signed amendments (if applicable) and, (3) 52.212-3 Completed and signed Representations and Certifications (Commercial Items). VOLUME 2: Technical Proposal (60points): The technical quotation should clearly and fully demonstrate the offeror's capabilities, knowledge and experience regarding the technical requirements described below. Failure to respond explicitly to each of the categories and subcategories listed below may result in the offeror's quotation being deemed technically unacceptable: Technical Capability: The proposed technical solution, shall include, but is not limited to the following: 1) A resume describing experience, 1-4 pages outlining education and experience and 2) Bibliography of authored publications ? contractor may list any publications they have. VOLUME 3: Past Performance 40 Points): The past performance shall demonstrate the contractor?s background and experience of the proposed personnel in the area of neuromuscular autoimmunity research. The contractor shall provide: 1) References: Offerors shall provide a minimum of two (2) references for who they have or are currently providing services similar to that described in the SOW. Price (Price will be evaluated but, will not be scored). The Evaluation Process: The Government will award a contract resulting from this quotation to the responsible contractor whose quote conforming to the quotation will be the most advantageous to the government, price and other factors considered. FAR 52.212-1, INSTRUCTIONS TO OFFERORS COMMERCIAL ITEMS and FAR 52.212-3, OFFEROR REPRESENTATIONS AND CERTIFICATIONS ? COMMERCIAL ITEMS, along with the DUNS NUMBER ADDENDUM. The resulting contract will incorporate FAR 52.212-4, CONTRACT TERMS AND CONDITIONS - COMMERCIAL ITEMS and FAR 52.212-5, CONTRACT TERMS AND CONDITIONS REQUIRED TO IMPLEMENT STATUTES OR EXECUTIVE ORDERS ? COMMERCIAL ITEMS. The following FAR Clauses cited in paragraph (b) of the clause at 52.212-5 are applicable to this acquisition.
