DESCRIPTION: A goal of managers in the FKNMS is to use aquaculture facilities to stabilize the health of corals rescued or salvaged from vessel groundings or construction sites. The eventual objective is to return some of these corals to their natural environment, while making some of the colonies available to scientists, public aquaria, and other educational and scientific purposes. The exchange or trade of protected species, such as corals, requires being able to determine the original source. In order to have a coral aquaculture program where protected species can be exchanged between scientists, public aquaria and others, and established mechanism for tracking a particular coral back to its parent colony is needed. This is particularly important in helping prevent the establishment of a coral black market. Corals that are propagated in aquaculture facilities from parent colonies that were collected under legal permits need to be tracked back to those parent colonies in order to prevent illegal trade of corals. DNA tracking has been used in marine mammals and other protected species but has not previously been attempted for tracking corals. If successful, this project will aid managers and other authorities in the management and control of the highly regulated corals used by aquaculture facilities. REQUIREMENTS: The FKNMS has a need to determine the feasibility of coral genetic fingerprinting as a mechanism to track native and cultured coral. Task 1 ? Determine the best method of DNA identification using various molecular techniques on one coral species. A gene specific identification of individuals, amplified fragment length polymorphisms, or other method may be used. Task 2 ? Produce a detailed report on each method with a recommendation for the preferred method for this purpose. Reports on each method will be due at 6 and 12 months for the life of the contract. Task 3 ? Provide a final report to FKNMS in digital and hardcopy formats. Report due annually at the end of each contract year. In addition to the above tasks if applicable, the option years will have the following tasks: Task 1a ? Using the best method as recommended in Task 1 or as recommended by FKNMS, determine DNA sequencing for other coral species. Task 2a ? Produce a detailed report for each species. Task 3a ? Provide a final, summary report to FKNMS in digital and hardcopy formats. Report due annually at the end of each contract year. Knowledge, Skills, and Abilities: Contractor must provide staff, equipment, and facilities for the above mentioned tasks. NOAA/FKNMS staff will provide contract related training when necessary. The contractor will provide all staff, equipment, and supplies necessary to perform the tasks described above. There are no benefits and the contractor is responsible for payroll taxes and insurance including medical. The contractor shall not be deemed a Federal employee for any purposes including Federal Tort Claims Act and Federal Employee Compensation Act purposes. Progress payments are authorized. Payments will be made via electronic funds transfer (EFT). NOTE: THE ABOVE ARE MIMUMUM REQUIREMENTS. BASE YEAR - BID FORMAT: Provide itemized pricing as follows: TOTAL Line Item #1 ?
