Description
The National Institutes of Health (NIH), National Cancer Institute (NCI), Center for Cancer Research (CCR) Genetics Branch (GB) intends to procure the Biotage Pyrosequencer PSQ HS 96 System SNP it has had in the laboratory for 10 months under a Leasing Agreement from Biotage Inc., 1725 Discovery Dr., Charlottesville, VA 22911. One of the GB studies is the identification and/or characterization of molecular markers that may be utilized in the early detection and prevention of cancer. The current emphasis is on the tumor-specific promoter region methylation of genes important in breast and prostate carcinogenesis. The Biotage HS SNP Pyrosequencer will be used in the quantitative assessment of the methylation status of many CpG sites simultaneously of genes important in prostate carcinogenesis. The Biotage PSQ Pyrosequencing HS 96 System SNP is based upon proprietary chemistry developed by Biotage/Pyrosequencing. The technology is a sequencing-by-synthesis method that relies on luminometric detection of pyrophosphate release upon nucleotide incorporation via an enzymatic cascade. The pyrosequencing technique offers a fast and efficient way to identify DNA (or cDNA) sequence variation and allows for the simultaneous and quantitative evaluation of variation in regions of up to 60 bases. The assays require a small amount of sample and have been shown to be able to quantify the degree of sequence variation with high accuracy. The PSQ Pyrosequencing HS 96 System SNP must be able to: 1) be able to determine gene methylation quantitatively down to 5% levels; 2) be able to determine quantitative methylation of multiple CpG sites simultaneously (at least 50 base pair regions); 3) be sensitive enough to determine PCR yields as low as 50 ng of product; 4) have a 96-plate format for high throughput analysis; and 5) have adequate software for conversion of data into Microsoft Excel. Biotage is the only known source to the NCI researcher that can provide a commercial piece of equipment that meets the aforementioned requirements. This is not a solicitation for competitive quotations. However, if any interested party believes it can meet the entire above requirement, they may submit a statement of capabilities. All information furnished shall be in writing and must contain sufficient detail to allow the NCI researcher to determine if it can meet the requirements of this synopsis. This acquisition will be processed under FAR Part 12 ? Acquisition for Commercial Items and will be made pursuant to the authority in FAR 13.5 to use simplified procedures for commercial acquisitions. An original and one copy of the capability statements must be received in the Contracting Office by 1PM EDT (local Washington, D.C. time) on July 22, 2005. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will be considered solely for the purpose of determining whether to conduct a competitive procurement.
