- In accordance with Simplified Acquisition Procedures the National Institutes of Health (NIH), National Cancer Institute (NCI), Center for Cancer Research (CCR), Medical Oncology Branch (MOB), Molecular Pharmacology Section (MPS), intends to procure on a sole-source basis with the University of Louisville, 2301 South Third Street, Louisville, KY 40292 for chemical synthesis of a number (n=8) of CC-5013 (Revimid) analogs, including EM-12. One of the objectives of MPS is to identify new agents to treat cancer, with a particular interest in prostate cancer. The compounds produced under this award will be used as reference standards for CC-5013 metabolism experiments. The modified compounds will be used to learn about the pathways of metabolism and hydrolysis of thalidomide. Since synthetic routes are not known for all of the compounds and will have to be designed before the synthesis can be performed. The University of Louisville shall: 1) Accurately develop and validate an synthetic route; 2) Produce 50 mg of ?d 95% pure HPLC grade raw compound (n=8) (structures will be provided for the eight compounds once the contract is awarded); 3) Accurately label and store samples in the freezer; 4) Accurately identify the sample throughout the process; 5) Fully characterize the compounds using MS and NMR and maintain the database of compounds in accurate, logical, and acceptable format; 6) Utilize good laboratory safety skills and handle all samples with universal precautions; 7) Utilize good laboratory techniques; 8) Provide full synthetic routes for each compound, including methods, yield and characterization data, to allow for replication and scale-up; and 9) deliver the eight raw compounds to the MPS within 2 months of award. Dr. Fred Luzzio at University of Louisville is to provide these services. They are a leader in the field of organic chemistry syntheses, with emphasis on thalidomide chemistry. They have extensive experience in the development of synthesis route for thalidomide analogs (CC5013 is an analog of thalidomide) and will be able to do the synthetic route development and scale up within a two month period of time. The University of Louisville is the only vendor known to the NCI Researcher that can meet the above requirements. This is not a solicitation for competitive quotations. However, if any interested party believes it can provide the above service, it may submit a statement of capabilities. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform the requirement. An original and one copy of the capability statements must be received in the contracting office by 1:00 p.m. EDT, on September 12, 2005. Faxed and emailed capability statements are NOT authorized. A determination by the Government not to compete this proposed requirement based upon responses to this notice is solely within the discretion of the Government.
