- It is the intent of the National Institutes of Health, National Eye Institute, to procure professional services from Dr. Michael K. Wyatt, of Annandale, Virginia, on a sole source basis. Period of performance will be one year from the date of award. This service is commercially available. The following information may be entered into a notice of intent synopsis for a noncompetitive (sole source) acquisition: 1. The contractor shall have specific expertise in the areas of functional genomics of critical interest to the program of the Section on Molecular Structure and Functional Genomics for an ongoing production of recombinant proteins for structure function studies. 2. The contractor shall have a B.S., M.S., and Ph.D. degree in biological disciplines, and shall have extensive experience in molecular biology as related to recombinant protein synthesis and expression. 3. The contractor shall have expertise and hands-on knowledge of yeast 2-hybrid analysis of functional interactions of eye expressed proteins, and shall have expertise in the ongoing research on the human RPE expressed targets of Complement Factor H, related to Age-related Macular Degeneration. 4. The contractor shall have knowledge of advanced techniques including random chimcragenesis and site specific mutagenesis. 5. The contractor shall have hands on experience with the Clontech MatchMaker system. 6. The contractor shall have specific experience in NIH?s facilities and a proven track record of production of eye-related proteins. 7. The contractor shall ensure that adequate supplies of recombinant proteins be produced efficiently to allow the program to proceed, and shall ensure that techniques for functional genomics, particularly Y2H to discover functional important interactions of proteins related to eye disease and development are exploited. 8. The contractor shall have experience in expression of recombinant proteins for functional and structural analyses, to include but shall not be limited to the following: Targets for CFH binding. CCP domains. Novel eye-expressed proteins including a klotho related protein that appears specifically to the eye. 9. The contractor shall have experience in mass spectroscopy, Polymerase Chain Reaction (PCR) of cDNA clones to generate recombinant constructs, subcloning into expression vectors, typically using the pET system, DNA sequencing to check validity of constructs, expression of recombinant protein, purification of proteins using Pharmacia AKTA FPLC system, and other relevant technologies. 10. The contractor shall identify specific targets for CFH interaction with RPE expressed proteins and supply of protein for several different experiments. 11. The contractor shall provide purified samples of recombinant proteins in sufficient quantity for specific experiments. 12. The contractor shall design and carry out molecular biology experiments, and shall follow up on the results of expression studies relevant to understanding functions of recombinant proteins and their possible connection to disease and important biological processes. These experiments shall include, but shall not be limited to the following: DNA sequencing. Polymerase chain reaction. Northern and Western blotting. Mass spectroscopy and functional assays. 13. The performance period shall be from April 14, 2006 through April 13, 2007. It is the Government?s belief that the above requirements can only be met by the Dr. Michael K. Wyatt. This is not a formal solicitation, however business concerns that respond to this notice must furnish concise responses directed specifically to the requirements mentioned above. Business concerns demonstrating the capability to accomplish the requirements shall supply pertinent information in the form of curriculum vitae in sufficient detail to demonstrate their ability to perform. Information furnished shall include enough material to perform a proper evaluation of each item listed above. Respondents will not be notified of the evaluation results.
