Description
This notice is a combined synopsis/solicitation for commercial items prepared in accordance with format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; quotes are being requested & a written solicitation will not be issued. Submit offers on RML-RFQ-6009. The solicitation documents and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2005-07 dated 02/02/06. This acquisition will be processed under Simplified Acquisition Procedures (SAP) and is a Small Business Set-Aside. The North American Industry Classification System (NAICS) code for this procurement is 339111 and the small business size is 500. The Rocky Mountain Laboratories requires a Confocal Microscope capable of extremely rapid image acquisition, continuous long-term imaging (over the course of hours and days, with greater than 300 images obtained) of living cells, very high sensitivity (single molecule detection), high spatial resolution (both axial [Z] and lateral [XY] dimensions) to accurately image fine cell surface structures, all without the need for image deconvolution. The system shall be capable of rapidly capturing (with at most 200 ms exposure times) both differential interference contrast (DIC) and fluorescence (multiple channels) images of a given field of view of weakly fluorescent specimens. Specifications for the system are: A fully motorized inverted microscope with the specifications as listed: a6-position filter turret with filter cube sets for DAPI, FITC, and Texas Red .Shall include built-in optical magnification tube of at least 1.5X. 5 (five) emission ports (both sides, eyepiece, front and bottom) to allow attachment of multiple cameras simultaneously. Shall include metal halide illumination system with variable intensity control for epi-fluorescent illumination (120 watt, 1500 hour rated, pre-aligned lamp with temperature monitor to prevent restrikes until lamp has cooled sufficiently and light guide to transmit light from source to microscope). Shall include accessories for DIC microscopy (prisms and analyzer), DIC objectives meeting specifications indicated: Plan apochromat 10X NA 0.45 WD 4.00 mm, Plan fluor 40X oil NA 1.3 WD 0.2 mm. Plan apochromat 60X water immersion NA 1.2 WD 0.27 mm (must have correction collar and chromatic aberration correction in near-UV range), Plan apochromat 60X oil TIRF NA 1.45 WD 0.13 mm (must have correction collar), Plan apochromat 100X oil TIRF NA 1.45 WD 0.13 mm. Internal Z-focus drive with high precision Z-axis automated focus with linear encoded Z-axis readout of 0.05 um minimum step. Shall include focus drift correction device to continuously monitor coverslip position with 770 nm wavelength LCD and immediately relay feedback to microscope focusing motor to maintain focal plane position over long periods of time with precision of less than 1/3 the focal depth of the objective. Shall include laser-based Total Internal Reflection Fluorescence (TIRF) illuminator with single molecule detection capability and clean-up filter sets (488, 568, and 643 nm). Shall include confocal system and associated components as listed: Scan head with at least 6 (six) user-selectable apertures consisting of at least 3 (three) slit modes (slits of 70 um, 35 um, and 20 um widths) and at least 3 pinhole modes (each consisting of arrays of pinholes of 45 um, 60 um, and 90 um widths) to permit complete optimization of sensitivity, speed, and resolution (both axial and lateral [X and Y]) for all imaging applications. Shall have motorized mechanism to change between different aperture settings. Scan head shall be capable of speeds up to 260 Hz in pinhole mode and 2600 Hz in slit mode. Shall have diode (442 nm) or air-cooled (488, 514, 568, 647 nm) lasers emitting lines at 442 nm, 488 nm, 514 nm, 568 nm, and 647 nm with acousto-optical tunable filter (AOTF) switching between lines. Interchangeable dichroic and barrier filters for 488/568 nm, 488/647 nm, 440/514 nm, and 488/568/647 nm (triple dichroic). Shall include imaging workstation PC with P4 3.06 GHz processor (or better) and at least one flat panel monitor. Shall include image acquisition and analysis software. Shall be capable of: driving all components of the microscope (including transmitted light source intensity) as well as external components listed in this document (e.g. cameras, Piezo motorized XYZ stage, emission filter wheel, etc.) with flexibility to add and control additional components. Shall include binary and 8-, 16-, 24-, 48-bit image display and processing, stack display and processing including best focus, montage, opographical surface, view orthogonal planes, and create movie. Shall include arithmetic operations including add, subtract, multiply, divide, logical AND, OR, XOR, minimum, maximum, and average. Shall include image processing manipulations such as shading correction and background subtraction, filters (low pass, median, unsharp mask). FRET/ratiometrics analysis, Time lapse acquisition, Image stitching, Image alignment, Colocalization and correlation measurements, Angiogenesis measurement, Count nuclei/cell scoring, Neurite outgrowth measurement, Motion analysis and particle counting and tracking in 3D (X,Y,Z or 2 dimensions plus time) and 4D (X,Y, and Z over time). Shall include 3D deconvolution with settings for confocal (scanning or spinning disc) or widefield images, multiple algorithms (blind, non-blind, and adaptive blind), correction for depth- attenuation/photobleaching. Shall include 2D deconvolution (with algorithms as for 3D deconvolution above). Shall include 3D reconstruction and visualization with real-time rendering and rotation of 3D data sets, isosurface viewer, easy 3D movie generation, image alignment. Shall include high sensitivity 16-bit back-illuminated EMCCD microscopy camera: 512 x 512 array of 16 x 16 um pixels, high quantum efficiency (>80% at all wavelengths between 450-750 nm), cooling to -80?C without water or liquid nitrogen (to minimize dark current to ~0.008 e-/p/s, thereby increasing sensitivity), user-selectable on-chip multiplication gain, up to 331 fps frame rates (measured at 10 MHz), selectable readout at 10, 5, and 1 MHz with on-chip gain available at 10 and 5 MHz. High resolution 16-bit back-illuminated EMCCD camera: 1004 x 1002 array of 8 um x 8 um pixels, good quantum efficiency (>50% at all wavelengths between 450-750 nm), low dark current (0.5 e-/p/s at -30?C), user-selectable on-chip multiplication gain, up to 103 fps frame rate (measured at 10 MHz). Shall include Piezo XYZ nanopositioning motorized stage (closed loop XY stage with rotary encoders) with controller and specimen holder for slides and small Petri dishes. Shall include 200 um Z travel distance, XY travel range of 110 mm x 90 mm, XY resolution of 0.088 um, Z resolution of 1.5 nm, and Z axis max velocity (with settling time) of 5 mm/sec. Shall include motorized high-speed, 8 position emission filter wheel with rapid switching (<40 ms between adjacent filters) with controller and mounting components plus emission filter sets (FITC/GFP, TRITC/Rhodamine, CY5, Thioflavin T). Shall include microscope incubation chamber (encloses entire microscope, not just stage top) with complete temperature, CO2 (with regulator to allow input of non pre-mixed gas), and humidity to maintain viable cells for long periods of time (days). Shall include anti-vibration air table, need dimensions of 30? x 42? by 35? tall to fit installation location. Quotes shall include on site installation and training by a licensed dealer complete with full manufacturer?s warranty and pricing on service contract (parts and labor) on all components.
