Description
NIST seeks information on commercially available mitochondrial DNA resequencing scanner systems. NIST may procure such systems after results of this market research are obtained and analyzed and specifications are developed after a determination is made regarding which mitochondrial DNA resequencing scanner systems can meet NIST's minimum requirements. The system shall be capable of the following: A scanner that can scan a chip tiled with DNA probes for the entire 16, 568 base pairs of DNA sequence of the mitochondrial genome in both the forward and reverse orientation and include redundant tiling of the 500 most common mitochondrial haplotypes. ●The scanner should read small feature size of 8 micrometers. Resolution should extend down to pixelation of 0.51 micrometers. The laser should have a sensitivity of 0.5 chromophore equivalents/micrometers squared or less. ●The software should be able to handle multiplex experiments and complete a single scan in 10 minutes. This will keep experiment turn-around time under 48 hours. The software should use adaptive background genotype calling scheme (ABACUS). The "adaptive" nature of this scheme should use hybridization data from multiple arrays across multiple runs to factor in the uneven background occurring due to differing levels of cross-hybridization at each site, and significantly reduce the occurrence of miscalls. ●Scanner should have capability for automated controlled sample loading. ●Scanner should support software that performs batch analysis. Once the batch analysis is completed, the software should generate a report containing individual and total numbers and percentages of base calls within the batch, and a detailed case-by-case list of genotype variations vis-?-vis the reference sequence. The software should analyze the data and list the mutations in an Excel format, with the exact position, the wildtype sequence and the base change displayed side by side for rapid comparison. ●The system should be widely validated for use on cancer clinical samples and should be able to operate in an environment running 10,000 scans per year. ●Laser Power ? Should be auto-set solid state green laser and reduce gain drift due to aging optics components. ●The following parameters have been used for NIST prior experiments and should be maintained in a scanner to ensure normalization of data. Excitation ? 532 nm (nanometer), 200?W Emission ? 570 nm, (nanometer) Long-Pass Detector ? Meshless Photomultiplier Tube, Red Enhanced Displayed/ Saved Dynamic Range ? 16-bit, (65535:1) Power: Voltage: 100-240V, Current: 4-2 A, Frequency: 50-60 Hertz ●The scanner size should similar to a desktop with dimensions of 13 inches W x 18 inches H x 22 inches deep. NIST is seeking responses from both large and small businesses. Small businesses are defined under the associated NAICS code for this effort, 334516, as those having 500 employees or less. Please include your size classification in any response to this notice. Companies that manufacture or sell commercially available mitochondrial DNA resequencing scanner systems are requested to email detailed specifications for such commercially available system(s) to todd.hill@nist.gov no later than the response date for this sources sought notice. Specifications shall include detailed functional and performance capabilities for the mitochondrial DNA resequencing scanner system. Also, the following information is requested to be provided as part of the response to this sources sought notice: 1. Name of the company that manufactures the people counting system for which specifications are provided. 2. Name of company(ies) that are authorized to sell the people counting system, their addresses, and a point of contact for the company (name, phone number, fax number and email address). 3. Indication of number of days, after receipt of order that is typical for delivery of such systems. 4. Indication of whether each instrument for which specifications are sent to todd.hill@nist.gov are currently on one or more GSA Federal Supply Schedule contracts and, if so, the GSA FSS contract number(s). 5. Any other relevant information that is not listed above which the Government should consider in developing its minimum specifications.
