Description
The USDA, Western Regional Research Center, Albany, California has a requirement for one (1) laser scanning confocal microscope, built around a DMI 6000 CS inverted bionocular base configured with four objectives: HC PL APO 20x/0.70 IMM CS, HCX PL FL L40x/0.60 fwd 3.3-1.9, HCX PL APO 40x/1.25-0.75 OIL CS, and HCX PL APO 63x/1.4-0.6 OIL CS. AOBS conventional scanner module, filter-less 4-channel spectral detection with a Red HeNE 633nm, Yellow DPSS 561nm, Blue Argon 458-514nm (includes exhaust and silencer). 405nm laser with power supply, ND filter wheel and UV optics. Includes a computer workstation with LAS AF SP5 offline dongle and 3d/4d processing software, microscope table with passive damping and panel box control of scanning parameters. System designed for future capability to be fully upgradeable with the addition of an independent tandem scanning head allowing rapid scanning capability useful in live cell imaging. Completely filter-free in the optical light path of the scanner. Fully computer-controlled acousto-optical beam splitter for any combinations of up to 8 laser excitation lines. Transmission of fluorescence emissions across visible wavelength of up to 90%. Suppression of Laser excitation lines to within 2nm of the wavelength used. Use of prism in spectral light path for highest transmission efficiency, up to 96%. Use of motorized slits in front of each spectral fluorescence detector. Fully computer-controlled for adjustment of the detection of the wavelength of fluorescence signal. Slits are controlled with nm accuracy and changeable during scanning for fine adjustment. Four spectral detectors, with water-cooled electronics and hardware integration to reduce image noise. Upgradeable to 5 spectral detectors. One transmitted light detector, usable simultaneously with all internal detectors. Scanner head provides exit port for access to confocal emission light path. Users can adapt/couple their own detection device to the port. Optional APDs (avalanche photo-diode) with 2 ? 4x higher quantum efficiency available for exit port attachment for low-light imaging. Scan rotation of at least 200 degree without effecting scan speed or optical resolution; sequential scan in line, frame and stack modes; true multiple region of interest (ROI) scan; imaging in 8/12/16 bit up to 8k x 8k pixel format; large scan field (FOV 22) with 10x objective, maximum field size of 1.5mm x 1.5mm; optical zoom up to 32x; single detection pinhole for perfect registration of confocal images from all detectors; 3 visible lasers (expandable to 8?UV/405-vis-IR) coupled to scanner head usable simultaneously; argon lasers (458nm, 476nm, 488nm, 496nm, 514nm), yellow DPSS laser (561nm), and red HeNe laser (633nm); all laser lines intensity controlled by acousto-optical tunable filter and freely combinable; 405nm laser, 50mW, usable in any combination with visible laser lines for imaging DAPI and Hoechst dyes; high-precision focusing stage, 1.5mm focusing range, 40nm minimum step size capable of high speed XZ live scanning; multi-functional digital panel box for direct control of scanning parameters; user programmable with LED display functions and sensitivity adjustments; spectral scanning of samples with 1nm minimum step size including spectral analysis software; fully integrated microscope with all operations from confocal software; all functions such as focus, filter change and objective change motorized.
