- REQUEST FOR INFORMATION SAIC-Frederick, Inc., on behalf of the National Cancer Institute?s Mammalian Gene Collection program, is requesting the following information from companies actively offering contract DNA synthesis: What would be the effect on the overall costs if two different clones for every cDNA ORF were synthesized, if the pairs of clones differ by a single base of the target? Specifically, for each assigned ORF, in addition to synthesizing one clone with its naturally occurring stop codon, what is the effect on cost in getting a second version of the construct which has the second base of the stop codon (of the same ORF) substituted with a ?C? (converting all three versions of stop codons to serine codons). Thus for the target gene, there would be two synthetic ORFs, each with and without a stop codon generated. What is the effect on the cost if 2 to 4 thousand cloned ORF sequences are targeted? Would the cost be impacted by the size of the construct or its GC content?
