- Description
- The objective of this contract is to provide NIEHS investigators with access to genetically altered mice. For transgenic studies, NIEHS investigators generate the transgenic construct by cloning the transgene of interest downstream of a tissue-specific or ubiquitous promoter. NIEHS investigators also develop genotyping methods to identify founder mice. The Contractor shall microinject the linearized transgenic constructs into single cell mouse embryos which are then implanted into pseudopregnant mice to produce founders. The Contractor shall assist with tail biopsies and genotyping, and deliver transgenic founder mice to NIEHS for expansion of the lines and phenotypic characterization. For knockout mice, NIEHS investigators generate the knockout construct and develop genotyping methods to identify targeted ES cell lines. The Contractor shall electroporate the ES cells with the construct, select and maintain the targeted ES cell lines, perform blastocyst microinjections, and perform implantations into pseudopregnant mice shall be compiled to produce chimeric offspring. The Contractor shall assist with tail biopsies and genotyping, and deliver chimera to NIEHS for expansion of the lines and phenotypic characterization. Detailed health surveillance information on all the mice prior to transfer to NIEHS. Examples of new transgenic and knockout mouse lines developed by NIEHS investigators over the past year under this contract include the following: pTETmITSN, Tie2-CYP2C8, Tie2-CYP2J2, N3-2, HABAf60a140, CMV-ITSN, pCIITIH2, pCIITIH5, tetolTIH2, tetolTIH5, rtTA, and rTS." The anticipated effective date for the award is September 30, 2007. The contract will consist of a two year base period and four two year options. The contract will be a cost reimbursement completion type contract. For estimating purposes the Government anticipates an approximate level-of-effort of 832 professional direct labor hours for the base period and 832 professional direct labor hours for each optional period for a total of 4,160 direct labor hours. All requests must reference RFP No. NIH-ES-07-03 and must be directed to the office listed above. All responsible sources may submit a proposal which shall be considered by the Agency.
