Description
The National Cancer Institute (NCI), Division of Cancer Epidemiology and Genetics (DCEG), plans to procure as a sole source for the brand name TECAN Infinite M200 Monochromator-based Modular Multi-function Microplate Reader and components. This acquisition will be processed in accordance with the simplified acquisition procedures as stated in FAR Part 13.106-1(b)(1). The North American Industry Classification System code is 334516 and the business size standard is 500 employees. JUSTIFICATION FOR OTHER THAN FULL AND OPEN COMPETITION: Part I: Background information and description of requirements: The overall goal of the NCI laboratory is to develop new approaches to the study of the genetic basis of cancer and its outcomes. Specifically, the NCI will investigate the genetic basis of strong association signals identified in candidate gene association studies, loci identified by linkage analyses in high-risk families, or genome-wide association studies (GWAS), particularly the loci identified by the ongoing Cancer Genetic Markers of Susceptibility (CGEMS) program. The goal of functional studies is to determine the mechanisms of gene function and how they are perturbed in disease. This research utilizes molecular biology, immunological, genetic and biochemical technologies. These include but is not limited to the following procedures: fine mapping of candidate loci, DNA and RNA quantification, protein quantification, reporter gene assays, cell viability and toxicity assays, immunoassays, enzyme assays and binding assays. Part II: Facts and Reasons to Justify Other than Full and Open Competition: Statutory Authority: 13.106-1 (b) and 13.501-(a)(1) (b) Soliciting from a single source. (1) For purchases not exceeding the simplified acquisition threshold, contracting officers may solicit from one source if the contracting officer determines that the circumstances of the contract action deem only one source reasonably available (e.g., urgency, exclusive licensing agreements, or industrial mobilization). Justification In order for the NCI laboratory to carry out functional studies of candidate genetic variants, we need the following capabilities: high-throughput UV and VIS absorbance, fluorescence top & bottom read with spectral emission scans in 1nm increments from 280 – 850nm, dual color luminescence measurements, dual injectors, and a cuvette port. The TECAN Infinite M200 Monochromator-based Modular Multi-function Microplate Reader is a single high-quality, easy-to-use instrument that incorporates the aforementioned detection methods needed to conduct our research. The Tecan Infinite M200 Microplate Reader is designed to address the widest range of microplate-based applications. It combines fluorescence filter-based and monochromator-based detection systems into a single unit. The filter-based system can be used for high sensitivity or fast read speeds and the flexible monochromator system can be used for spectral scanning. It also has a quadruple monochrometer tunable in 1 nm steps for access to all wavelengths from UV to NIR to perform absorbance, excitation, and emission scans. The quadruple monochrometer provides more efficient blocking of undesired wavelengths for more accurate readings. Further, this is the only instrument that also includes: a spectrally enhanced photomultiplier tube, a luminescence module capable of reading dual color, a dual injector, temperature control, and a patented NanoQuant plateTM for high-throughput DNA & RNA concentration measurements with only 2 ul of sample. Detailed Instrument Requirements: -Detection Modes: Fluorescence Intensity (FI) Top/FI Bottom/Absorbance (Abs) Luminescence Photon Counting (Lumi) -Wavelength Selection: Quadruple monchrometer (tunable in 1nm steps); 2-Excitation (EX) and 2-Emmission (EM) monochrometers -Bandwith Selection: EX: 5nm for λ ≤ 295nm; 9nm for λ>295nm; EM: 20nm -Light Source: 10-Watt UV Xenon Flash Lamp -Detectors: Photomultiplier Tube (PMT): analogue-type for fluorescence measurements; Standard PMT: EX 230-600 nm, EM 330-600nm; Optional spectrally enhanced PMT: EX 230-850nm, EM 280-850nm; UV Silicon Photodiode for absorbance measurements; Photon Counting (CPS) PMT for luminescence -Measurement Speed: 20 sec, 96-well plate; 30 sec 384-well plate; Wavelength EX/EM-scan, 96-well plate, 450-550nm, 5nm step: 125 sec -Plate formats: 6,12, 24, 48, 96 and 384-well plates; cuvettes. User created plate definition files in software that allow for custom microplate definitions for all plate types. -Software: i-Control – standard (raw data acquisition placed in MS-Excel©); Magellan (instrument control and full data reduction) -Cuvette Port Module: 10nm path length -Temperature Incubation: Ambient +5ºC up to 42 ºC -Shaking: Linear and orbital; 1-600 secs; amplitude from 1 – 6mm in 0.5 mm increments -Fluorescence Measurements: a.Wavelength range: 230 – 280nm in 1nm increments b.Measurement range: 0-60,000 RFU/0-6,000,000 RFU (extended dynamic range) c.PMT Gain Adjustment: 1 – 255 d.FI top reading: 1fmol/well e.FI bottom reading: 9fmol/well f.TRF Integration times adjustable for 20-2000μsec in 1 μsec steps -Absorbance Module: g.Wavelength Range: 230-1000nm h.260/280 nm measurements i.Measure 16 samples simultaneously ii.Separate quartz optic for each sample iii.Sample volume ≥ 2μl iv.Sensitivity: 1 ng/μl v.Compatible with multichannel pipettes -Luminescence Module: i.Wavelength Range: 400-700nm j.Linear Dynamic Range: 8 decades k.Cross-talk (96-well plate): <0.01% l.Sensitivity: 1fmol ATP/well m.Dual color: BRET assay capable (green & magenta filters installed) -Injector Module: n.Pump speed: 100 – 300 μl/sec o.Injection volume: Selectable in 1μl increments; minimum volume 5μl; maximum volume 800μl per stroke p.Dead volume: 100μl including pump back This notice is not a request for competitive quotation. However, if any interested party believes it can meet the above requirement, it may submit a statement of capabilities. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform the requirement. No faxed or emailed capability statements will be accepted. Contractor shall provide one (1) original and one (1) copy of the capability statement. Capability statements must be received in the contracting office by 1:00 PM EST, on June 20, 2008. A determination by the Government not to compete this proposed requirement based upon responses to this notice is solely within the discretion of the Government. Information received will be considered solely for the purpose of determining whether to conduct a competitive procurement. No collect calls will be accepted. In order to receive an award, contractors must be registered and have valid certification in the Central Contractor Registration (CCR) and the Online Representations and Certifications Applications (ORCA).
