Description
The National Cancer Institute (NCI), Division of Cancer Epidemiology and Genetics (DCEG) plans to enter into a sole source procurement for the nested case-control study in the Prostate, Lung, Colorectal, and Ovarian (PLCO) Cancer Screening Trial from Johns Hopkins University, Sidney Kimmel Comprehensive Cancer Center, Cancer Research Building, Room 1M53; 1650 Orleans Street; Baltimore, MD 21231. The supplies and services herein are being procured in accordance with the simplified acquisition procedures authorized by FAR Part 13. The North American Industry Classification System Code is 541712 and the business size standard is $7 million. The purpose of this procurement is to quantify genomic 5-methyl-2’-deoxycytidine (5meC) content in DNA samples of (1) 40 blinded duplicate pilot samples, and (2) 500 colorectal cancer cases, 500 matched controls, and 10% blinded duplicate quality control samples, using a validated, sensitive, high throughput method. Recent studies have measured global DNA methylation levels in genomic DNA using peripheral blood, and correlated hypomethylation status with increased cancer risk (e.g. colorectal adenoma, head and neck cancer, bladder cancer) as well as carcinogen exposure (e.g. low dose benzene). Furthermore, hypomethylation has been associated with folate deficiency among cancer patients and healthy controls, consistent with animal studies. Evidence also exists to suggest a relation of global methylation status with smoking, alcohol, HPV infection, and other environmental factors. The National Cancer Institute (NCI) conducts a study to evaluate genomic global DNA hypomethylation in relation to colorectal cancer risk and its potential interrelationship with folate deficiency and other risk factors. It is important to confirm or refute previous case-control findings using prediagnostic samples such as those collected in the PLCO Trial. Period of performance: Period of performance: (1) 40 pilot samples: September 1, 2008 through September 30, 2008; (2) 1100 study samples: September 25, 2008 through April 15, 2009 At the present time, no other laboratory is equivalent for reasons of assay validity, cost and DNA resource preservation. Therefore, Johns Hopkins University is the sole source which meets these requirements. The method proposed here, High Pressure Liquid Chromatography/ Tandem Mass Spectrometry, was developed and validated at the Johns Hopkins University. The approach developed at Johns Hopkins University is cost-effective, because it is high throughput with relatively low labor requirements (i.e. 30-40 study samples per run in half day work of one FTE). The approach requires only 1% of the DNA needed for other methods (10 ng of genomic DNA). This is not a solicitation for competitive quotations. However, if any interested party believes it can perform the requirement as detailed they may submit a statement of capabilities. All information furnished shall be in writing and must contain sufficient detail to allow the NCI to determine if it can meet the above unique specifications described herein. An original and one copy of the capability statement must be received in the contracting office by 1:00 pm EDT, on August 29, 2008. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will be considered solely for the purpose of determining whether to conduct a competitive procurement.
