Description
The National Cancer Institute (NCI), a division of The National Institutes of Health (NIH) plans to procure on a sole source basis analysis of telomere samples with Dr. Immaculata DeVivo Associate Professor at Harvard Medical School, 181 Longwood Avenue Boston, MA 02115. The supplies and services herein are being procured in accordance with the simplified acquisition procedures as stated in FAR Part 13.106-1(b)(1). The North American Industry Classification code is 622110 and the business size standard is 31.5M employees. Period of Performance: One year from date of award. The primary objective of this procurement is to determine telomere length in a nested case-control study of gastric cancer cases and controls. Although a large number of studies have evaluated the role of telomere length in tumor biology and the aging process, a smaller number of studies have investigated the role it may play as a risk factor for cancer. Limited data are available on the impact of telomere length and telomerase activity in the gastric mucosa. Experimental studies have observed increased telomerase activity and decreased telomere length in gastric tumor tissue compared to normal gastric tissue. The pathway from exposure to shortened telomere length and subsequent elevated cancer risk has not been well examined, particularly for gastric cancer for which no evaluation of germline telomere length has been conducted to date. In this study, the contractor will propose to measure germline telomere length by Q-PCR of genomic DNA in gastric cancer cases and controls from a pooled nested case-control study. Cases and controls will be drawn from four prospective studies: the Shanghai Women’s Health Study (SWHS), the Multiethnic Cohort Study (MEC), the Prostate, Lung, Colorectal and Ovarian Cancer Screening Trial (PLCO), and the Women’s Health Initiative (WHI). The purpose of this study is to investigate whether telomere length is associated with gastric cancer risk and to relate measures of chronic inflammation and oxidative stress with telomere length. Existing and study generated data on H. pylori status and virulence, gastric atrophy, markers of chronic inflammation and oxidative stress from prospectively collected specimens will be used. Knowledge gained from this study will provide important information on characterizing telomere length and its relation to gastric cancer. In order to allow direct comparison of data from all studies, it is necessary for telomere length Q-PCR measurement to be performed in the same laboratory as the previous study of this procurement. (Reference Contract # MQ703153) The purpose of the current procurement is to determine telomere length by high-throughput Q-PCR in 450 gastric cancer cases, 900 controls, and an additional 150 blinded quality control samples. Contractor Requirements: The contractor shall perform the following tasks: 1.Analyze average telomere length in 1500 samples in triplicate using a Q- PCR assay (e.g. total 4500 analyses). 2.Provide a plan for determining if the replicate values are in agreement and for repeat duplicates, as needed (allow 5% sample replenishment). 3.The dates of service will extend from August 18, 2008 through August 17, 2009. The proposed contractor, Brigham and Women’s Hospital of Harvard Medical School, has an operating telomere assay that is capable of measuring average telomere length in DNA using limited amounts of DNA and with high-throughput to analyze the needed number of samples. The other techniques available (Southern blot, FISH) require large amounts of DNA that are not available within these studies and lack high-throughput capabilities rendering these techniques unsuitable for the proposed studies. The requested scope of the work can only be reasonably achieved with this novel high-throughput assay, as alternative low-throughput methods like Southern blots would take a fulltime analyst several years to complete. Therefore, due to the urgency of the research and the lengthy time requirement, it has been determined Brigham and Women’s Hospital laboratory is the only known source to the NCI which can meet all of the Governments requirements. This is not a request for competitive quotation. However, if any interested party believes it can perform the above requirement, it may submit a statement of capabilities. The statement of capabilities and any other furnished information must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform this requirement. An original and one copy of the capability statements must be received in the contracting office by 11 a.m. ET August 29, 2008. Faxed and E-mailed capability statements are not authorized. A determination by the Government not to compete this proposed requirement based upon responses to this notice is solely within the discretion of the Government.
