- The National Cancer Institute (NCI) plans to procure on a sole source basis, Small RNA Libraries for High Throughput Screening from Systems Biosciences, Inc; 1616 N. Shoreline Blvd; Mountain View, CA 94043. This acquisition will be processed under FAR Part 12 – Acquisition for Commercial Items and will be made pursuant to the authority in FAR 13.106-1(b)(2) and 13.501-(a)(1) to use simplified procedures for commercial acquisitions. The North American Industry Classification System Code is 541712 and the business size standard is 500 employees. Large-scale RNA interference (RNAi) studies have facilitated the search for genes required for diverse biological processes enabling stepwise dissection of specific signaling pathways. In combination with high-throughput assays, genome-wide RNAi studies have uncovered novel gene functions in various biological processes through loss of function approaches. Several of these studies were aimed at the identification of genes essential for cell division, cell cycle progression, endocytosis, tumor transformation and apoptosis. Through these studies, the power of this approach for the identification of functional modules has been demonstrated. The Center for Cancer Research has an immediate requirement to obtain state-of-the-art tools for applying high-throughput, RNAi-mediated gene silencing to study various pathways and processes in mammalian cell systems. A combination of short hairpin RNA vector libraries in both viral (lentivirus) and plasmid forms are being sought to support these types of loss-of-function assays. In addition, microRNA precursor pooled libraries are also being sought to assist the Center for Cancer Research (CCR) investigators in the understanding of the role of miRNAs in biological processes, such as in cancer development. This library will enable the study of phenotypic effects associated with the over-expression of individual microRNAs. The required features of the libraries include: <!--[if !supportLists]--> 1) <!--[endif]-->Pooled stocks in both virus (lentivirus) and plasmid form; <!--[if !supportLists]--> 2) <!--[endif]-->A demonstrated history of success through a record of publications AND a minimum of three years of commercial availability; <!--[if !supportLists]--> 3) <!--[endif]-->High complexity libraries of a minimum of 160,000 shRNA sequences in a single pool, allowing genome-wide coverage in a single screen; <!--[if !supportLists]--> 4) <!--[endif]-->The use of bio-safe lentivectors that have been demonstrated to provide superior transduction efficiency. These viruses must be able to infect difficult-to-transduce cells, including hematopoietic stem cells, with high efficiency (>80). <!--[if !supportLists]--> 5) <!--[endif]-->Lentivectors and lentiviral packaging tools to enable high titer viral preparations, with >10 9 IFUs/mL concentration possible; <!--[if !supportLists]--> 6) <!--[endif]-->Data Analysis support for genome-wide libraries to assist with interpreting results of high-throughput RNAi screens; <!--[if !supportLists]--> 7) <!--[endif]-->Libraries must be in-stock and available for immediate shipment of over 10^8 IFUs of each packaged Library or 1 mg of Plasmid Library stock; <!--[if !supportLists]--> 8) <!--[endif]--> Identification of enriched shRNAs can be performed through hybridization to a standard Affymetrix chip; and <!--[if !supportLists]--> 9) <!--[endif]--> A prepackaged microRNA library representing the majority of known microRNAs (>500 individual miRNAs). Genome wide shRNA screens have proven to be extremely valuable for cancer research. The CCR Center of Excellence in Integrative Cancer Biology and Genomics believes that access to this technology must be made available to members of the CCR as soon as possible and in a manner that is logistically reasonable. Libraries developed by Systems Biosciences, Inc., meet all of the requirements of the CCR. JUSTIFICATION: 1. Statutory authority: Statute: 13.106-(b)(2) and 13.501(a)(1) (b) Soliciting from a single source. (2) For Sole source acquisition of commercial items in excess of the simplified acquisition threshold conducted pursuant to subpart 13.5, the requirements at 13.501(a) apply. Systems Biosciences, Inc. is the only known vendor to the NCI that can fulfill all of the requirements of this solicitation as outlined above. This is not a request for competitive quotation. However, if any interested party believes it can meet the above requirement, it may submit a statement of capabilities. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow the researcher to determine if the party can meet this requirement. One (1) original and one (1) copy of the capability statement must be received in the contracting office by 1:00 p.m. ET on September 24, 2008. A determination by the Government not to compete this proposed requirement based upon responses to this notice is solely within the discretion of the Government. In order to receive an award, contractor must have valid, updated registration and certification in the Central Contractor Registration (CCR) and the Online Representations and Certifications Application (ORCA).
