Description
The National Cancer Institute (NCI),Genetic Epidemiology Branch, plans to procure on a sole source basis with deCODE Genetics, 1000 Winter St. Suite 3100, Waltham, MA 02451. The contractor will perform genotyping with microsatellite markers on genomic DNA samples extracted from blood collected from 1,500 participants in nasopharyngeal carcinoma families. The North American Industry Classification System Code is 541711 and the business size standard in 500 employees. This acquisition will be processed in accordance with simplified acquisition procedures as stated in FAR Part 13.106-1(b)(1). The objective of this procurement is to conduct fine-mapping by genotyping additional microsatellite markers in the regions showing suggestive linkage signals in the genome-wide scan. Specifically, genomic DNA from 1500 individuals in about 200 Nasopharyngeal Carcinoma (NPC) multiplex families will be genotyped for deCODE fine mapping markers at 2 cM interval. The results are required in order to confirm or exclude linkage regions that showed linkage signals in the initial genome-wide scan. These data will be critical in narrowing the regions for candidate gene sequencing. The vendor shall provide: 1.Genotyping data 2.Map locations for markers genotyped 3.Marker allele frequencies 4. Mendelian inconsistency check Contractor Requirements: 1.Design and validate microsatellite markers in the regions that NCI provides; 2.Provide 2-ml screw-cap tubes for the DNA, boxes labeled with order ID, and barcode labels for tubes. 3.Check whether the DNA is enough for 40 STR fine mapping and at the required concentration; 4.Conduct extensive QC before, during, and after genotyping according to deCODE procedure. QC before genotyping includes OD measurements, normalization and a panel of 50 STR on all samples to see that they all perform; 5.Conduct genotyping and include appropriate CEPH individuals as quality controls; 6.Deliver genotyping results and map files; 7.Provide technical guidance and trouble shooting to the NCI Project Officer on the work performed under this procurement. deCODE Genetic's genotyping service delivers rapid-turnaround, high-quality data for linkage analyses. The company operates the largest and most advanced high-throughput microsatellite and SNP array genotyping facility in the world. Backed by years of experience, deCODE has created the most complete genetic map of the human genome and an unrivalled track record in mapping and validating disease genes for common diseases. deCODE has successfully genotyped 360 individuals for ~1000 microsatellite markers in the genome-wide scan for melanoma family study in DCEG. Their service was fast and had great quality, with high allele call rate and extremely low Mendelian inconsistencies. For fine mapping, deCODE has over 15,000 validated markers that successfully have been used for genotyping. The well validated marker set required to follow-up on the two regions of interest to our study are uniquely available through deCODE, therefore justifying the need to this procurement. This is not a request for competitive quotation. However, if any interested party believes it can meet the above requirement, it may submit a statement of capabilities. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow the researcher to determine if the party can perform this requirement. An original and one copy of the capability statement must be received in the contracting office on or before 11:00 am ET on April 21, 2009 to Melissa Marino, Contract Specialist. A determination by the Government not to compete this proposed requirement based upon responses to this notice is solely within the discretion of the Government.
