- The National Cancer Institute (NCI), Center for Cancer Research (CCR), Experimental Immunology Branch (EIB) plans to procure on sole source basis: A) Special Order Laser Upgrades of an existing BD Biosciences FACSAria II Flow Cytometer/Cell Sorter and; B) Replacement of existing obsolete equipment with a Special Order 6-Laser BD Biosciences FACSAria II Flow Cytometer/Cell Sorter with BD Biosciences, 2350 Qume Drive, San Jose, CA 95131. This acquisition will be processed under FAR Part 12 - Acquisition for Commercial Items and will be made pursuant to the authority in FAR 13.106-1(b)(2) and 13.501-(a)(1) to use simplified procedures for commercial acquisitions. The North American Industry Classification System Code is 334516 and the business size standard is 500 employees. Only one award will be made as a result of this solicitation. The objective of the acquisition is to provide two equipment upgrades in the Flow Cytometry Core Facility of the Experimental Immunology Branch (EIB), CCR, NCI: 1.) Special Order Laser Upgrades of an existing BD Biosciences FACSAria II Flow Cytometer/Cell Sorter and 2) Replacement of existing obsolete equipment with a Special Order 6-Laser BD Biosciences FACSAria II Flow Cytometer/Cell Sorter. The EIB is proposing to procure the Laser Upgrade of BD FACSAria SN# P22300031 from BD Biosciences because, as the Original Equipment Manufacturer (OEM), BD Biosciences is the only known vendor that can perform these modifications on an existing BD Biosciences instrument and meet the specifications as described below. This requisition is for extensive modifications to an existing instrument to upgrade the laser delivery system of that instrument. The upgrade involves replacing the existing lasers on the instrument with higher power lasers that are completely compatible with the fiber optics laser light delivery and fluorescent light collect systems in the instrument. This optical system is a fundamental component of the instrument. Only the OEM can perform the upgrade in a way that maintains full integration with OEM optical system and the OEM computer control hardware and software. Only the OEM can provide the upgrade in a way that provides for complete maintenance coverage of the entire system by the OEM. (Specifications stated below.) The EIB is proposing to procure the new 6-Laser Flow Cytometer/Cell Sorter from BD Biosciences because The BD Biosciences FACSAria II flow cytometer/cell sorter is the only known available system that provides all the analytical and cell sorting capabilities as described below. Further, the BD FACSAria II has several unique capabilities required for support of existing research projects in our core facility. Finally, only the BD FACSAria II provides for uninterrupted continuity of research and immediate compatibility with existing software that is under development. BD Biosciences has 5 local service engineers trained on the BD FACSAria II, more than any other known vendor. This capability is required for minimizing down time and is critically important due to the nature of the work in the EIB core facility. SPECIFICATIONS Special Order Laser Upgrades of an existing BD Biosciences FACSAria II 1. Provide and install ARIA SPECIAL ORDER LASER SYSTEM on existing BD FACSAria SN# P22300031. 2. To include installation of fiber launched 100mw 488nm Coherent Sapphire laser, 40mw 640nm coherent Cube laser and a 50mw 405nm violet Point source laser (all measurements at the laser head). These three new lasers will replace the existing three lasers in the ARIA flow cytometer. 3. Existing optics to be used. 4. Installation to include hardware/software necessary to provide operator control of laser power output for all three new lasers. 5. All components installed to be compatible with existing optical systems and computer control systems. 6. To include full warranty/maintenance coverage under OEM warranty and maintenance agreements. 7. Complete on-site (Bld. 10, Room 6B16) installation. 8. Upgrade to be compatible with existing and future maintenance coverage by BD Biosciences of BD FACSAria SN# P22300031. BD Biosciences FACSAria II SPECIAL ORDER SYSTEM To consist of: 9. 6 lasers and 18 associated fluorescence detectors as follows: 488nm laser (50mw) with forward scatter, side scatter, and five fluorescence detectors 640nm laser (40mw) with three fluorescence detectors 561nm laser (50mw) with three fluorescence detectors 594nm laser (200mw) with three fluorescence detectors 405nm laser (100mw) with three fluorescence detectors 355nm laser (100mw) with two fluorescence detectors To include the ability to use all 18 fluorescence and 2 scatter detectors simultaneously for both data collection and for cell sorting. 10. The 561nm and the 594nm lasers will be colinear and share the same detectors. 11. The 640nm and the 594nm lasers to have high performance detectors to optimize red fluorescence detection 12. Option for laser power monitoring and control software for all lasers. 13. High speed cell sorting (>24,000 cells/sec) capability that uses a gel-coupled cuvette detection system and octagon and trigon based fluorescence collection systems fiber coupled from collection lens. To include automated software control for set up and adjustment of drop delay during sorting. To include automated software for self regulation during sorting that provides ability to shut down sample and sheath flow, and to cover sorted materials in presence of gross shift in sorting parameters. 14. Two-way and four-way high speed cell sorting capabilities to sort into microtubes, 12x75mm tubes and 15 ml tubes, including sample agitation and temperature control for both unsorted sample and sorted cells. 15. Automated Cell Deposition Unit (ACDU) to allow sorting into multiwell plates or onto microscope slide. The ACDU cloning unit shall have capability to select unique sort regions for different wells within the same 96 well plate. 16. Optical filters to detect the following fluorophores: FITC, PE, PE-TxRd, PerCP-Cy5.5, PE-Cy7, APC, Alexa700, APC-Cy7, Alexa594, mRasberry, AQ143, mCherry, DsRed, Tomato, Pacific Blue, AmCyan, Qdot605, DAPI/Hoechst, Indo-1, Side Population Filter Set 17. Computer Hardware and FACS DIVA Software necessary for instrument control and data collection and including TWO LCD display monitors. 18. Temperature Control Option and Aerosol Management Option The temperature control capabilities shall include ability to cool samples both pre and post sort, including sorted cells in either tubes or 96 well plates. 19. OEM supplied Instrument Table and Computer Table 20. To include digital electronics (from the preamp), automated digital electronic compensation between all laser and all fluorescence parameters (including algorithms for calculating digital multi-color compensation across all laser channels), the ability to real time ratio any signal from any laser, the ability to collect Height, Area and Width parameters on all parameters simultaneously, and baseline restoration algorithm. To include computer system for digital data collection that can output data file compatible with FCS version 2 and FCS version 3 file standards. 21. Must have fixed alignment lasers that do not require operator alignment or focus. 22. Must be installed and serviced by in-state service personnel with 48 hour response time guaranteed. 23. Must include 1 year full parts and on-site labor warranty to cover all malfunctions except consumables. This is not a solicitation for competitive quotations. However, if any interested party believes they can meet the above requirement, they may submit a statement of capabilities. All information furnished must be in writing and must contain sufficient detail to allow the NCI to determine if it can meet the above unique specifications described herein. An original and one copy of the capability statement must be received in the NCI contracting office on or before 11:00 AM EST on July 27, 2008. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government.
