Description
The National Institutes of Health (NIH), National Cancer Institute (NCI), Center for Cancer Research (CCR), Laboratory of Tumor Immunology and Biology (LTIB) intends to procure on a sole-source basis a BD LSR II laser upgrade with BD Biosciences, 2350 Qume Dr., San Jose, CA 95131. This acquisition will be processed in accordance with simplified acquisition procedures as stated in FAR Part 13.106-(b)(1). The North American Industry Classification System Code is 334516 and the business size standard is 500 employees. The Laboratory of Tumor Immunology and Biology is a large translational research laboratory that conducts research in the areas of tumor immunology, mechanisms of tumor cell-immune cell interactions, and immune mechanisms. The laboratory functions as an integrated translational research program with the goal of designing and developing new immunotherapies and immunologic strategies for cancer treatment and prevention. One crucial piece of equipment used by multiple investigators is the BD Biosciences LSRII flow cytometer. The flow cytometer is used by many investigators and provides crucial experimental data on the expression of antigen by a variety of cells as well as their functional characteristics. The data is important in the overall assessment of the efficacy of different vaccine modalities to elicit a host immune response. The current LSR II flow cytometer in LTIB has two lasers (blue laser/488nm and red laser/633nm). There is a limitation of only 6-color flow cytometric analysis with the two lasers. The laboratory analyzes patient samples pre and post vaccination for monitoring of specific immune responses. More than 6-color will be needed for the phenotypic analysis of markers. Additional lasers (violet laser and UV laser) are needed for the use of additional fluorochromes (Pacific blue, AmCyan, Marine blue, Alexa Fluor 405, Cascade blue, Cascade yellow, CEP and Alexa Fluor 430) and DNA analysis as well as calcium flux for the functional analysis of T effector and Treg cells. Multiple markers are needed for the identification of specific subpopulations of T cells, such as central and effector memory, naïve T cells, and memory and naïve Tregs. Current configuration of our LSR II will not allow LTIB to perform this study. Additional color capability will allow LTIB to study the subpopulations that are responding or not responding to the vaccine treatment. Addition of these lasers to the current LSR II flow cytometer will facilitate the clinical monitoring of patients immune responses due to vaccination and the generation of more valuable information for the design of clinical trials. Similar studies are also ongoing using mouse models in LTIB. Additional lasers will greatly facilitate the mouse model studies as well. The proposed upgrade will add functionality to the existing FACSLSRII flow cytometer by increasing the number of features and areas of interest on each cell type under investigation. The current configuration of the instrument allows for study of six dyes on each cell simultaneously. The violet laser upgrade will add four additional colors that can be detected. Software will also be included. This upgrade will consist of a 405nm laser, a 355nm laser, electronics to power the lasers and optics for laser beam induced cell signal detection. The upgrade is only available direct from BD Biosciences, who is the original manufacturer of the LSRII cytometer. This upgrade is integrated into nearly every system of the cytometer which is the reason it is available only from the manufacturer. BD Biosciences, manufacturer of the FACSLSRII flow cytometer sells both instruments and upgrades directly to the end user and does not use distributors. For these reasons, the upgrade needs to be purchased from BD Biosciences as there is no other known source. This is not a solicitation for competitive quotations. However, if any interested party believes it can meet the above requirement, they may submit a statement of capabilities. All information furnished shall be in writing and must contain sufficient detail to allow the NCI to determine if it can meet the above unique specifications described herein. An original and one copy of the capability statement must be received in the NCI contracting office by 1:00 PM EDT on August 4, 2009. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will be considered solely for the purpose of determining whether to conduct a competitive procurement.
