- The National Cancer Institute (NCI), Division of Cancer Epidemiology and Genetics (DCEG), plans to procure on a sole source basis Protein Staining of Colorectal Tumor Tissue Microarrays (TMA’s) Constructed in the Prostate, Lung, Colorectal, and Ovarian (PLCO) Cancer Screening Trial from the University of California at Los Angeles, Department of Pathology & Laboratory Medicine; Reed Neurological Research Center, Rm 3155; 710 Westwood Blvd; Los Angeles, CA 90095. This acquisition will be processed in accordance with simplified acquisition procedures as stated in FAR Part 13.106-1 (b)(1). The North American Industry Classification System code is 621511 and the business size standard is $13.5 million. The period of performance shall be twelve (12) months from date of award. Currently, NCI is conducting a study to evaluate the associations of risk factors with subtypes of colorectal carcinoma and adenoma, as determined by tumor protein staining status of P53, APC, Cyclin D1, EGFR, and VEGF. TMA’s provide a high-throughput approach to analyzing multiple protein expression in situ on large series of patient samples, but may be limited by the pathologist’s ability to reproducibly score large quantity of samples and markers, distinguishing subtle differences in staining intensity using a continuous scale, and accurately scoring ratio of protein expression between subcellular compartments (e.g., ratio of signals from the nucleus and the cytoplasm). The NCI will evaluate the automated Aperio ScanScope scoring system in a pilot study by comparing its results with those by traditional pathologist manual review of the immunohistochemistry (IHC)-stained TMA slides. Upon satisfactory completion of the pilot evaluation, the full study will be implemented by the chosen scoring method to determine protein staining status of P53, APC, Cyclin D1, EGFR, and VEGF for 14 colorectal carcinoma TMAs and 5 adenoma TMAs. The proposed contractor will carry out the laboratory analysis to measure protein staining status of colorectal tumors, including acquisition of optimal reagents, optimization of staining, pilot test runs, and scoring across all array spots, totaling 4,935 tissue spots in 19 tissue microarrays per marker. The Department of Pathology and Laboratory Medicine, UCLA, under the direction of Dr. David Seligson, collected PLCO tissue cores, constructed the TMAs, established the test array of colorectal tumors for the pilot study, and provided satisfactory staining data for phase I of the study where a pilot testing of four proteins were successfully conducted. Dr. Seligson’s laboratory is the repository for PLCO Trial pathology tumor arrays; all the samples and associated TMA data are physically stored at UCLA; thus, no other source has these or suitably similar materials to conduct the study. This is not a request for competitive quotation. However, if any interested party believes it can meet the above requirement, it may submit a statement of capabilities. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow the researcher to determine if the party can meet this requirement. One (1) original and one (1) copy of the capability statement must be received in the contracting office by 11: 00 a.m. EST on September 3, 2009. A determination by the Government not to compete this proposed requirement based upon responses to this notice is solely within the discretion of the Government.
