Description The National Cancer Institute (NCI), Laboratory of Cellular and Molecular Biology (LCMB) plans to procure the brand name or equal Sorvall MTX-150 Benchtop micro-ultracentrifuge and S120-AT3 Fixed Angle Titanium Rotor, 14 x 0.5ml. This acquisition will be processed in accordance with simplified acquisition procedures as stated in FAR Part 13.106-1(b)(1). Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. The Laboratory of Cellular and Molecular Biology (LCMB) studies the molecular basis of cancer cell migration, invasion and metastasis. The focus of the lab is to understand the molecular basis for the catalytic activity, regulation and cellular function of the Arf GAPs with the ultimate goal of developing therapeutics targeting specific Arf GAPs. In current work, the LCMB found that (1) the interaction of Arf GAPs with membrane surfaces is critical to their function (2) the Arf GAPs function in protein complexes on membrane surfaces and (3) each Arf GAP has distinctive structural requirements for catalytic activity and regulation. The ultracentrifuge is necessary for studying the interaction with membrane surfaces, including large unilamellar vesicles (LUVs) and purified Golgi apparati, in small volume assays. It is also necessary for the preparation of LUVs used in many assays including assays of protein-protein interaction for those associations that depend on a surface. The Sorvall MTX 150 Micro-Ultracentrifuge are engineered to deliver performance and versatility. The MTX 150 is the smallest unit in its class yet achieves the highest speeds (150,000 rpm) and the highest g-force (1,048,000 x g) with rotor capacities of up to 180ml. Quiet operations (<48 dBA) and standard electrical outlet capabilities have the added convenience of being able to place the unit anywhere in the lab. The salient characteristics for the required benchtop micro-ultracentrifuge are as follows: 1) A footprint of no more than 2.3 square ft. 2.) The top speed of 150,000rpm. 3.) The unit operate at 48dBA which makes it comfortable to quietly run in the lab. 4.) Rotors that can spin 6 X 30ml volumes and a swinging bucket that holds 4 X 7ml volumes.
