- The National Heart, Lung and Blood Institute (NHLBI) Office of Acquisitions (OA) has a requirement for the National Dental and Craniofacial Research (NIDCR), The Division of Intramural Research (DIR) for consulting services to produce Recombinant AAV Vectors and test for neutralizing antibodies in serum samples. The reference solicitation number is: NHLBI-PB-(DE)-2010-099-DDC. BACKGROUND INFORMATION: Sjogren’s Syndrome is a systemic, chronic autoimmune disease of unknown ideology which affects 1-3million Americans primarily affecting the lacrimal and salivary glands. Patients complain of dry eyes and dry mouth the latter leading to pain, discomfort, dental carries, and infection of the mouth by opportunistic pathogens. Women are 9 times more likely than men to be affected in the general population. There are currently no affective therapies for treating this disease. PURPOSE AND OBJECTIVES OF THE PROCUREMENT: The Molecular Physiology and Therapeutics Branch (MPTB), Division of Intramural Research (DIR), National Institute of Dental and Craniofacial Research (NIDCR) requires the services of a consultant with expertise in performing adeno-associated virus (AAV) vector production and test for neutralizing antibodies in serum samples. This expertise is necessary in order to expand upon our current studies in patients suffering from Sjögren’s Syndrome. Primary Sjögren’s Syndrome (SS) is a systemic autoimmune disorder characterized by exocrine gland dysfunction primarily involving the salivary and lacrimal gland. The histologic hallmark of Sjögren’s Syndrome is a periductal inflammatory infiltrate. The conventional explanation of salivary gland dysfunction assumes a causal relationship between this chronic inflammatory process and the decreased saliva production. However, most patients have preserved salivary gland tissue and in overall the correlation between the focus score on SG biopsy (a measure of inflammation) and saliva production is limited. Recent studies have suggested that some immunomodulatory proteins may have beneficial effects in animal models of the disease and warrant further testing. CONTRACTOR REQUIREMENTS: The specific tasks to be performed are to prepare and analyze samples from Sjogren’s patients or test animals for the presence of neutralizing antibodies to different serotypes of AAV vectors and to produce recombinant stocks of vectors. The Contractor shall research data, including results obtained through the testing process, as well as other relevant patient data. The Contractor is responsible for the collection and the maintenance on a daily basis in the laboratory of Molecular Physiology and Therapeutics Branch (MPTB) on site in Building 10. Reporting Requirements: The completed final summary must be submitted within thirty days after the ending of the contract, and all reports shall be submitted electronically to the Project Officer. The estimated level of effort for this requirement is not expected to exceed 744 hours during the period of performance, for the period March 22, 2010 through September 30, 2010. The Contractor will have knowledge and experience in specific tasks as in preparing and analyzing serum samples from Sjogren’s patients or testing animals for the presence of neutralizing antibodies to different serotypes of AAv vectors by using antibody neutralization assays, this is a FACS based antibody neutralization system. A qualified candidate for this position should have at least 8 yrs experience performing AAV vector production as well as experience with Adenovirus vector production, cell culture, experience with testing for virus neutralizing antibodies, and a publication record in peer reviewed journals. Technical Evaluation Criteria: 1. Demonstrated knowledge and understanding of the NIDCR Sjogren’s Syndrome program with specific knowledge of serotypes, promoter, and transgene combination for studies covered under NIDCR protocol 84-D0056 and 94-D-0018. 2.Knowledgeable in specific tasks when preparing and analyzing samples. 3.Experienced in performing AAV vector production as well as experience with Adenovirus vector production. The North American Industry Classification (NAICS) Code is 541712 and the business size standard is 500 employees. This is a small business set-aside. This acquisition is being conducted using Simplified Acquisition Procedures in accordance with FAR Part 13. Proposals are due March 15, 2010 by 7:30 a.m. local time. The award will be made based on the lowest priced technically acceptable quote meeting the Government’s requirements. The Government intends to evaluate offers and award a contract without discussions with Offerors. Therefore, the initial offer should contain the offeror’s best terms from a technical and price standpoint. However, the Government reserves the right to conduct discussions if later it is determined by the Contracting Officer to be necessary. The Government may reject any or all offers, waive informalities and minor irregularities in offers received. The offeror’s proposal’s must include an itemized labor hour rates, complete description of services, complete address for invoice payment and purchase order delivery, prompt payment discount terms, the Dun & Bradstreet Number (DUNS), the taxpayer Identification Number (TIN) and certification of business size of the company. The offeror must also register in the Government’s Contractor Registry System. Note: www.ccr.gov. The proposal must reference “Solicitation number” NHLBI-PB-(DE)-2010-099-DDC.
