Description
THIS IS A NOTICE OF INTENT, NOT A REQUEST FOR PROPOSALS. A SOLICITATION DOCUMENT WILL NOT BE ISSUED AND PROPOSALS WILL NOT BE REQUESTED. It is the intent of the National Heart, Lung, and Blood Institute (NHLBI), for the National Human Genome Research Institute (NHGRI), to negotiate/award a purchase order to Sigma-Aldrich, 3050 Spruce Street, St. Louis, MO, 63103, for the design and testing of custom zinc finger nuclease. Sigma-Aldrich currently holds exclusive commercial rights to generate zinc-finger nucleases to target specific genes in Zebrafish. The technology was initially developed by Sangamo Biosciences and has been licensed to Sigma-Aldrich for sale to the research community. NHGRI Zebrafish Core provides services to NHGRI researchers that facilitate analysis of genes involved in human genetic diseases using zebrafish as a model system. To this effect, NHGRI has been performing targeted re-sequencing of ENU-mutagenized males (TILLING) for identification of mutations. This technology is limited by the number of males screened and the size of the exons as ENU generates mutations throughout the genome randomly. Therefore, for genes where TILLING has not identified knockout mutations, zinc-finger nuclease technology can be used for specific targeting. Therefore, Zebrafish Core is ordering zinc-finger nucleases for 2 genes. Contractor Requirements: -Sigma- Aldrich will perform the following tasks: -Design ZFN pairs for genes identified by the Core -Assemble ZFP modules -Attach DNA-cleaving Nuclease -Transfect in a cell line to validate designed ZFN candidates by mismatch sensitive nuclease assay -Prepare mRNA from the ZFN pair with highest DNA-cleavage efficiency -Provide plasmids and mRNA from validated ZFN pairs to the Core. Government Responsibilities: NHGRI will provide Sigma-Aldrich with information on two target genes. Sigma-Aldrich will keep NHGRI informed of progress on design, assembly and validation phases until the products are delivered to us. This acquisition is being conducted under FAR Part 13, Simplified Acquisition Procedures, and is exempt from the requirements of FAR Part 6, Competition Requirements. The North American Industry Classification System (NAICS) Code is 325188 and the Size Standard is 100 Employees. This notice of intent is not a request for competitive proposals. The determination by the Government not to compete the proposed purchase order based upon responses to this notice is solely within the discretion of the Government.
