Description
THIS IS A NOTICE OF INTENT, NOT A REQUEST FOR PROPOSALS. A SOLICITATION DOCUMENT WILL NOT BE ISSUED AND PROPOSALS WILL NOT BE REQUESTED. It is the intent of the National Heart, Lung, and Blood Institute (NHLBI) for the National Human Genome Research Institute (NHGRI), NIH Chemical Genomics Center (NCGC), to negotiate/award a purchase order to Applied Biosystems LLC, 850 Lincoln Centre Drive, Foster City, CA 94404, for the purchase of an Ambion Silencer Mouse Genome siRNA Library. The Trans-NIH RNAi initiative conducts high-throughput siRNA screens in collaboration with intramural investigators. Over the past year, NCGC have completed a series of successful pilot screening campaigns. These screens employed siRNA libraries targeting more than 7,000 human genes (roughly one third of the genome). Screening revealed novel components of important cell signaling pathways and genes that affect the activity of therapeutic agents. With the success and promise of these efforts, as well as the Initiative’s recent increase in throughput capacity, NCGC now requires a genome-wide mouse siRNA library to help fulfill our mission. Specifically, NCGC requires a library that •incorporates features to maximize gene silencing •incorporates features that minimize off-target effects •builds on our existing experience with siRNA technologies •targets more than 17,000 mouse genes •offers the library in a 384 well format that is broken into subsets, including the druggable genome •offers the library in an arrayed format presenting a single siRNA sequence per well •provides enough reagent to perform more than 100 genome-wide screens. The Ambion Silencer Mouse Genome siRNA Library meets all of these requirements. Several aspects of Ambion Silencer Mouse Genome siRNA Library make it unique. Silencer siRNA Libraries feature combinations of Silencer Pre-designed siRNAs and Silencer Validated siRNAs. The design of each siRNA has been carefully optimized to provide potent and specific silencing. The siRNA selection process yields siRNAs that are effective at low concentrations. Most importantly, this is the only genome-wide mouse library on the market that is offered in a single siRNA per well, 3 siRNA per gene format. Screening with a single siRNAs per well allows for increased confidence in hits whereby more than one siRNA against a given gene exhibits activity. One of the most critical aspects to successful siRNA experiments is maximal knockdown of intended target genes with limited off-target effects. This is imperative as off-target effects can lead to misleading phenotypes and greatly enhance that rate of false positives. To ensure specificity, the Ambion siRNA design algorithm incorporates a rigorous check of all siRNA sequences against the transcriptome. This process is accomplished via a modified alignment search tool that effectively identifies short regions of sequence alignment missed by BLAST searches. Sequences that have significant regions of sequence similarity to off-target transcripts are eliminated. Also eliminated are sequences that fall in polymorphic regions (e.g., SNPs) and sequences predicted to invoke an antiviral response. In addition, the siRNA design process incorporates stringent rules to prevent off-strand targeting. The sole source determination is based on the fact that Applied Biosysytems is the sole manufacturer and distributer of the Ambion Silencer Mouse Genome siRNA Library and no other product will fulfill the Government’s need. Product Description: Mouse genome 384 well 0.25 nm SiRNA Lib V3 This acquisition is being conducted under FAR Part 13, Simplified Acquisition Procedures, and is exempt from the requirements of FAR Part 6, Competition Requirements. The North American Industry Classification (NAICS) Code is 334516 and the Size Standard is 500 Employees. This notice of intent is not a request for competitive proposals. The determination by the Government not to compete the proposed purchase order based upon responses to this notice is solely within the discretion of the Government.
