- THIS IS A NOTICE OF INTENT, NOT A REQUEST FOR PROPOSALS. A SOLICITATION DOCUMENT WILL NOT BE ISSUED AND PROPOSALS WILL NOT BE REQUESTED. It is the intent of the National Institutes of Health (NIH), National Heart, Lung, and Blood Institute on behalf of the National Human Genome Research Institute (NHGRI), NIH Chemical Genomics Center (NCGC) to negotiate/award a purchase order to Tecan U.S. Inc, P.O. Box 13953, Research Triangle Park, NC 27709, for the purchase of an Infinite M100 Quadruple Monochromator Microplate Reader. The reference number is NHLBI-PB-HG-2010270-RSF. The Tecan M1000 is needed to facilitate NCGC’s assay development and technology evaluation program. This program is to provide extramural and intramural researchers with access to assay development support, as well as allow the Laboratory to evaluate novel assay formats. NCGC will work as a collaborative resource for doing assay development and validation screening. The Tecan Infinite M1000 will be used for a wide variety of applications for which it is suited, ranging from ratiometric, multiplexed reporter assays, to the characterization of chemical and natural product extract libraries at various stages of purification. It will also be used to determine possible interference in any qHTS assay that library is run against, and counterscreens to verify such interactions. The Tecan Safire Microplate Spectrometer Reader, a predecessor to the Infinite M1000, has been and is in current use extensively at NCGC for compound library characterization, as well as assays. These established protocols would need to be adapted and re-assessed if moved to another platform, as well as requiring the in-house expertise that already exists to require training on a different platform. Additionally, the extensive data collected and analyzed using methods developed with Magellan data analysis software and formats would need to be built from the ground-up for a different system, and possibly recollected on a different platform. As an example, in-house software we spent considerable time to develop and optimize to find the extinction coefficients of uv-vis absorbance maxima from 1536-well compound library plates collected on the Tecan platform would need to be re-written, if possible, to accommodate a different data format. The Infinite M1000 is unique because it can access a variety of microplate experiments within a single instrument. It can perform multiwavelength experiments, such as assays using dual fluorescent labels or two-color luminescent reporters, as well as collect full 2D UV, visible, near-IR, and 2D and 3D fluorescence spectra. The Tecan Infinite M1000 uses a monochromator-based detection system that eliminates the need for a collection of optical filters that need to be changed when changing assay detection wavelengths, so for example, assay plates with different dual-fluorescent label systems, can be studied back-to-back without the need to aquire different filter sets. No other type of supply will satisfy agency need. The determination by the Government not to compete the proposed purchase order based upon responses to this notice is solely within the discretion of the Government.
