Description
JUSTIFICATION FOR OTHER THAN FULL AND OPEN COMPETITION. Contracting Activity:USDA-ARS St Paul, Minnesota intends to Award as a Sole Source to Illumina. Nature/Description of Action: Request for purchasing laboratory consumables for conducting genotyping experiments using the high throughput genotyping assay method developed by Illumina. Description of Supplies/Service: The chemical reagents included in the high density bead chip kit custom designed for conducting genotyping assays, known as Infinium assay, manufactured by Illumina. Statutory Authority: In accordance with 41 U.S.C. 253(c) and FAR 6.302-1. Demonstration of Contractor's Unique Qualifications: A high density genotyping assay, known as the Infinium assay, developed by Illumina allows thousands of different DNA markers to be analyzed in 96 individuals in a single reaction assay, thus, permitting a large amount of data to be generated in a short period of time. The USDA-ARS laboratory in Fargo, ND has been using Illumina's reagents to perform similar genotyping experiments since 2006 with success. A set of 9000 wheat markers has been previously custom designed, and the Fargo lab has performed experiments using this set of markers on 1000 wheat samples so far with excellent results. We plan to use this set of 9000 wheat markers to genotype additional 384 wheat genotypes. The re-order of this marker set included in the high density bead chip kit along with other chemical reagents are manufactured by Illumina. Illumina is the sole manufacturer of the reagents. The equipment necessary to perform these assays is already available in the Fargo lab. The Fargo lab has already invested large amount of funds (~$300K) to equip our lab for conducting these assays. The Fargo lab requires no training on performing the Infinium assay system. Moreover, using the same assay system and marker design will provide us with the data consistency and allow cross comparison among different experiments generated at different time in the lab. Determination of Fair and Reasonable Costs: The Illumina genotyping technique is designed for high throughput genotyping experiments. The item stated in #3 will be used in the Fargo lab to conduct experiments and evaluated in the Illumina equipment to produce data in a large-scale. The cost for each assay using this method is at $60/sample, or less than one cent per data point, which is a very reasonable cost. Market Survey:The item stated above is manufactured and custom made at Illumina. Illumina developed the Infinium assay method and, thus, is the sole manufacturer of these chemical reagents for the assay, and no substitute vendor is available.
