Description The National Cancer Institute (NCI), Division of Cancer Epidemiology and Genetics (DCEG), Laboratory of Transitional Genetics plans to procure on a sole source basis Applied Biosystems BigDye Terminator v3.1 cycle sequencing kits from Applied Biosystems of 850 Lincoln Center Dr Foster City, CA 94404. This acquisition will be processed in accordance with simplified acquisition procedures as stated in FAR 13.106-1(b)(1). The North American Industry Classification System Code is 325413 and the business size standard is 500 employees. Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. The goal of NCI, DCEG, Laboratory of Translational Genomics is to dissect the genetic basis of single nucleotide polymorphisms (SNPs) validated in large scale, genome-wide association studies (GWAS). To accomplish this goal, thousands of patient DNA samples are sequenced each year to identify common gene variants, known as single nucleotide polymorphisms (SNPs). This genotyping is done using a well-established, high-throughput sequencing platform that utilizes BigDye Terminator v3.1 (BDT) cycle sequencing chemistry. BDT has been used for over nine years in to provide high-accuracy base assignments for comparative sequencing applications, namely SNP identification. The Laboratory of Translational Genomics receives various types of DNA extracts from all over the world that require sequencing, so it is imperative that the sequencing chemistry performs well on a wide range of template types and qualities. In order to continue the laboratory's mission of validating genetic variants that have been found to be associated with increased risk for certain cancers in GWAS, more cycle sequencing reagent with the following specifications is required: • The reagent must be a ready reaction mix containing dye terminators, deoxynucleoside triphosphates (dNTPs), Taq DNA polymerase, magnesium chloride and reaction buffer for quality control. • The reagent is required to be compatible with the current sequencing platform, 3730xl DNA Analyzer from Applied Biosystems. • The dyes in the reagent must be compatible with "Sequence Analysis Software version 5.2". • The dyes in the reagent must be compatible with "Data Collection Software" version 3.0. • The reagent needs to be formulated to generate long reads, as opposed to short, rapid read sequences. • The reagent must be flexible to perform well a variety of templates including PCR products, plasmids and large insert clones. • The reagent is required to be robust to perform well with difficult templates such as those with dinucleotide repeats and other challenging sequence motifs. Applied Biosystems BigDye Terminator v3.1 cycle sequencing kit is compatible with the existing sequencing platform, Applied Biosystems 3730xl DNA Analyzer, and the dyes in the reagent are also compatible with the equipment's software; Sequence Analysis Software version 5.2 and Data Collection Software version 3.0. AB BigDye Terminator v3.1 cycle sequencing kit is the only known reagent that satisfies the requirements. This is not a solicitation for competitive quotations. However, if any interested party believes they can meet the above requirement, they may submit a statement of capabilities. All information furnished must be in writing and must contain sufficient detail to allow the NCI to determine if it can meet the above unique specifications described herein. An original and one copy of the capability statement must be received in the NCI Office of Acquisitions on or before 11:00 AM EST on August 16, 2011. No electronic capability statements will be accepted (i.e. email or fax); an original and one copy must be sent to the NCI Office of Acquisitions to the address stated above. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government.
