- The USDA-ARS, on behalf of the Genomics and Bioinformatics Research Unit (GBRU), Stoneville MS, intends to negotiate and award on a non-competitive basis purchase order to Applied Biosystems, 850 Lincoln Centre Drive, Foster City, CA, 94404-1128 for the purchase of a BigDye Terminator v3.1 Cycle Sequencing Kit constituting 5000 reactions using 8 ml per reaction. Thus ~ 40,000 ml of Big Dye terminator v3.1 will be procured that has sufficient quality standard to produce a sequencing reaction of a 5 ml reaction using 0.25 ml of Big Dye terminator v3.1 on an ABI 3730XL DNA analyzer. The BigDye Terminator v3.1 Cycle Sequencing Kit's robust, highly flexible chemistry is ideal for de novo, resequencing and finishing with PCR product, Plasmid, Fosmid, and Bac Template. In operations at GBRU, a normal reaction consists of 0.25 microliters of BigDye mix in a 5 microliters sequencing reaction to receive a Phred 20 Quality score of 1000 on plasmid DNA and with 1.0 microliter on BAC DNA. The BigDye 3.1 is needed for ongoing projects within GBRU. It is a specialized mixture of a modified Taq DNA polymerase for cycle sequencing with florescent labeled ddNTPs that are detectable on an ABI 3730XL sequencer. The ABI 3730XL in the MSA Genomics Laboratory are under service contract and any material used cannot invalidate said warranty. GBRU supports research projects involve DNA analysis of a wide range of species important to agriculture. This reagent is vital to the success of GBRU related projects and has been validated on existing sequencing systems. Sequencing laboratory methods are necessary to determine these genetic variants. Delivery will be at Stoneville, Mississippi. Delivery is 30 days after receipt of order. These kits are proprietary to Applied Biosystems and are not available through another known source. The acquisition is being conducted under simplified acquisition using FAR Part 12 procedures and exempt from the requirement of FAR Part 6, Competition Requirement. This notice of intent is not a request for competitive proposals. Interested parties may identify their interest and capabilities in response to this synopsis. The determination by the Government not to compete the proposed contract based upon responses to this notice is solely with the discretion of the Government. The Government will consider all responses received by 4:00PM central time, Aug. 19, 2011. Information received will be considered solely for determining whether to conduct a competitive procurement.
