Set Aside: n/a NOTICE TEXT: The United States Army Medical Acquisition Activity (USAMRAA) intends to negotiate, on a sole source basis, with Promega Corporation, 2800 Woods Hollow Road, Madison WI 53711-5399, and award an Indefinite Delivery/Indefinite Quantity Requirements Type Contract to include a base year + four (4) one-year option periods to extend services for the purchase of DNA reagents. The Armed Forces DNA Identification Laboratory is an American Society of Crime Laboratory Directors accredited forensic lab. With the accreditations strict guidance to adhere strictly to established Quality Assurance (QA) and Standard Operating Procedure (SOP) to include consistency and continuity of test procedures. After validating a product, it is set to be used exclusively throughout the forensic identification. Validation and using various products is not possible from this point because it violates the consistency and continuity of obtaining accurate test results. Under the authority of FAR 6.302-1 (a)(2)(ii), only one responsible source and no other supplies or services will satisfy agency requirements. The estimated value of this requisition, including the four one-year option periods is $8,385,460.00. This is not a formal solicitation. This synopsis is not considered a Request for Quotation or Proposal. No solicitation document is available and telephone requests will not be honored. No contract will be award on the basis of offers received in response to this notice. Any response to this notice must show clear and convincing evidence that competition would be advantageous to the Government. If no affirmative written response is received, the purchase will be awarded without further notice. A determination by the Government not to compete this proposed contract based on this notice is solely within the discretion of the Government. The items to be incorporated into the contract will consist of: A - PowerPlex 16 (DC6530): this is a multiplex STR system for use in DNA typing, including paternity testing, forensic DNA analysis, human identity testing and tissue culture strain identification. The system should allow co-amplification with three-color detection of sixteen loci (fifteen STR loci and Amelogenin): Penta E, D18S51, D21S11, TH01, D3S1358, FGA, TPOX, D8S1179, vWA, Amelogenin, Penta D, CSF1PO, D16S539, D7S820, D13S317 and D5S818. One primer for each of the Penta E, D18S51, D21S11, TH01 and D3S1358 loci should be labeled with fluorescein (FL); one primer for each of the FGA, TPOX, D8S1179, vWA and Amelogenin loci should be labeled with carboxy-tetramethylrhodamine (TMR). One primer for each of the Penta D, CSF1PO, D16S539, D7S820, D13S317 and D5S818 loci should be labeled with 6-carboxy-4', 5'-dichloro-2', 7'-dimethoxy-fluorescein (JOE). All sixteen loci should be amplified simultaneously in a single tube and analyzed in a single injection or gel lane. The PP16 should be compatible with the Applied Biosystems 3130, 3130xl, and 3500 Genetic Analyzers that are currently used in the laboratory. B - The PowerPlex 16 HS (DC2100) (PP16 HS) shall allow co-amplification with three-color detection of sixteen loci (fifteen STR loci and Amelogenin), including Penta E, D18S51, D21S11, TH01, D3S1358, FGA, TPOX, D8S1179, vWA, Amelogenin, Penta D, CSF1PO, D16S539, D7S820, D13S317 and D5S818. One primer for each of the Penta E, D18S51, D21S11, TH01 and D3S1358 loci should be labeled with fluorescein (FL); one primer for each of the FGA, TPOX, D8S1179, vWA and Amelogenin loci should be labeled with carboxy-tetramethylrhodamine (TMR). One primer for each of the Penta D, CSF1PO, D16S539, D7S820, D13S317 and D5S818 loci should be labeled with 6-carboxy-4',5'-dichloro-2',7'-dimethoxy-fluorescein(JOE). All sixteen loci should be amplified simultaneously in a single tube and analyzed in a single injection. The PP16 HS should provide all of the materials necessary for amplification of these loci, including hot-start Taq DNA polymerase, and should be compatible with the Applied Biosystems 3130 and 3130xl Genetic Analyzers that are currently used in the laboratory. C- The PowerPlex 18D (DC1802,DC1808) in 200 reactions and 800 reactions should allow co-amplification with four-color fluorescent detection of eighteen loci, including D3S1358, TH01, D21S11, D18S51, Penta E, D5S818, D13S317, D7S820, D16S539, CSF1PO, Penta D, Amelogenin, vWA, D8S1179, TPOX, FGA, D19S433 and D2S1338. The PP18D shall provide all materials necessary to amplify STR regions of genomic DNA, including a hot-start, nonproofreading thermostable DNA polymerase. The system should be optimized for analysis of common database samples, such as unwashed FTA card punches (i.e., direct amplification). Additionally, thermal cycling should be reduced from previous PowerPlex protocols. The PP 18D system should be compatible with the Applied Biosystems 3130, 3130xl, 3500 and 3500xL Genetic Analyzers currently used in the laboratory D - The DNA IQ system (DC6700) should be designed specifically for the forensic and paternity communities. This system shall easily and efficiently prepare DNA for use in mtDNA and short tandem repeat (STR) analysis, eliminate PCR inhibitors, and remove contaminants frequently encountered in casework samples. The Elution Buffer (A8281) and the Lysis Buffer (A8261) are components of this system. Together with their chemistries, the DNA IQ system should be capable of extracting various sample types including buccal swabs, liquid blood, stains on FTA and other blood cards. With the addition of a pretreatment from supplementary kit such as the Tissue and Hair Extraction Kit (for use with DNA IQ), it shall be capable of extraction from additional sample types such as hair, or fresh, frozen, formalin-fixed, and paraffin-embedded tissue. The system shall deliver a consistent amount of extracted DNA, 1 to 4ng/ l in a 100 l elution, and be void harmful reagents requiring special disposal. E- 9948 Male DNA (DD2061) The male DNA standard shall be derived from male cell line 9948 and be supplied at a concentration of 10ng/ul. F- Tissue and Hair Extraction Kit (DC6740) is a supplementary kit used for the DNA IQ system. It shall contain an incubation buffer, lyophilized Proteinase K with specific activity of 34mAu/mg. G- DTT, Molecular Grade (V3155) is an antioxidant that should stabilize enzymes and other proteins containing sulfhydryl groups. The liquid form of the product is a 100mM solution of DTT in water. H - The DNA IQ Casework and Reference Sample Kit (AS1040) for Maxwell 16 AS1040 is used with Maxwell 16 instrument. These should provide and easy method for efficient, automated purification of genomic DNA from forensic and paternity reference samples. I- The Internal Lane Standard 600 (DG1071) should consist of 22 bands ranging in size from 60bp to 600bp. Fragments of 60-200bp are spaced at 20bp intervals, fragments of 200-500bp are spaced every 25 bases, and fragments of 500-600bp are spaced every 50 bases. J - Power Plex Matrix Standards (DG4650) should contain 4-dye matrix standards labeled for four fluorescent dyes. These standards should allow the Power Plex 16 (PP16) and Power Plex Hot Shot (PPHS) to be analyzed on the Applied Biosystems Genetic analyzers 3100/3130.
