Description
This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; price quotations are being requested and a written solicitation will not be issued. This solicitation number is RFQ2858119 and the solicitation is issued as a Request for Quotation (RFQ). The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2005-62. The National Institute on Drug Abuse (NIDA), Station Support/Simplified Acquisitions (SS/SA) Branch, has a requirement on behalf of the National Institute of Neurological Disorders and Stroke (NINDS), to generate Fis1 conditional knock out mice for the Surgical Neurology Branch in NINDS. This acquisition is associated with studies aimed at deciphering the function of Fis1, a mitochondrial protein controversially thought to be involved in mitochondrial dynamics. In order to conduct these studies, NINDS requires a contractor to provide an animal model knocking out Fis1 to confirm findings that we have cell culture and extend them into normal and differentiated tissues including neurons and muscle. The results shall be delivered to the National Institute of Neurological Disorders and Stroke (NINDS), 49 Convent Dr., Building 49, Room 2D-19, Bethesda, MD, 20892. The period of performance is ten months from date of award. The contractor shall provide the following: 1.The cloning and construction of the conditional knockout vector to flox exons 2-4 of the Fis1 gene with loxP sites with the additional insertion of FRT sites. Deliverables: vector. 2.Vector DNA must be purified and linearized. 3.The linearized targeting vector will be electroporated into C57Bl/6J ES cells and those cells will be placed under drug selection. Drug-resistant clones will be picked. 4.Clones will be screened with either long range PCR or Southern blotting to identify a portion of the recombinant Fis1 construct i.e. targeting the presence of a homologous arm. Clones will then be screened for the presence of the other arm. Deliverables: ES clones. 5.ES cell clones will be injected into blastocysts and the blastocysts subsequently injected into pseudo-pregnant mice to obtain chimeras. 6.The floxed Fis1 chimeric mice will be bred to FLP deleter mice on a C57Bl/6J background to obtain germline F1 pups with the drug-selection cassette removed. Deliverables: floxed Fis1 chimeric mice, F1 generation floxed Fis1 mice without neo cassette. The contractor shall provide a guarantee that if germline transmission of the desired construct cannot be achieved, NINDS will receive full reimbursement for funds invested into the project. Price quotations will be evaluated on meeting technical specifications and price. Offerors must include a completed copy of the provision at 52.212-3, Offeror Representations and Certifications Commercial Items, with its offer.
