Description
This is a combined synopsis / solicitation for Real-Time Quantitative Polymerase Chain Reaction (QPCR) machine. QPCR machine for gene expression analyses Real-Time Quantitative Polymerase Chain Reaction (QPCR) machine in order to continue on-going, time-sensitive studies as part of the Deepwater Horizon Natural Resources Damage Assessment.Studies on the impacts of the Deepwater Horizon oil spill to early life history stages of pelagic fish in the Gulf of Mexico. Over the last two years, researchers at NWFSC have developed or are in the process of developing QPCR assays for measuring the impacts of the crude oil exposure on fish embryos and larvae. These data were collected on an aging Applied Biosystems ABI 7700 QPCR machine that recently failed but is no longer supported by the manufacturer due to its age (~15 years), and cannot be repaired. Services to be provided: 1. The use of QPCR technology allows the measurement of indicators of cardiotoxic injury resulting from exposure to crude oil. 2. A replacement QPCR machine is needed that will allow for direct comparison between past and future results, thereby retaining data continuity and reducing costs associated with new reagents and labor for repeat optimization of assays, redesign of data analysis working protocols, and other QA/QC measures. 3. The replacement must be capable of running both 96- and 384-well blocks and "fast cycling" to reduce the time required for one assay from 2hrs to 45 minutes. 4. The replacement also must have adequate sensitivity to detect small changes in expression (> 1.5-fold). Evaluation Criteria: We will evaluate on the QPCR having the following capabilities: • Has capability to use previously purchased ABI reagents (e.g. master mix) and consumables (e.g. plates) • Has capability of running assays optimized on ABI 7700 or ABI 7900HT • Has capability to collected data continuous with data previously collected on ABI 7700 or ABI 7900HT (i.e., continuous with Analysis of Data protocols) • Has enhanced fluorescence detection • Has easily interchangeable thermal cycling block formats (96- and 384-well) • Has "fast" 96- and 384-well block capabilities • Has capability to detect 6 excitation (450-670nm) and 6 emission (500-720nm) wavelengths • Has multiplexing capabilities • Has multiple ramp method detection formats 1 each $___________________ Privacy Work on this contract may require that personnel have access to Privacy information. Personnel shall adhere to the Privacy Act, Title 5 of the U.S. Code, Section 552a and applicable agency rules and regulations. Data may not be released without COTR's written consent. Data must be protected from unauthorized disclosure. Contractor must comply with all Federal, DOC, NOAA, and NMFS policies relating to IT and data security. This procurement is being procured in accordance with FAR Part 13, Simplified Acquisition Procedures.
