INTRODUCTION: THIS IS A PRE-SOLICITATION NON-COMPETITIVE (NOTICE OF INTENT) SYNOPSIS TO AWARD A CONTRACT WITHOUT PROVIDING FOR FULL AND OPEN COMPETITION. The National Institute on Drug Abuse (NIDA), Station Support Contracts and Simplified Acquisitions Branch (SS/SA) on behalf of the NIDA intends to negotiate and award a contract without providing for full and open competition to AB Sciex LLC for the AB SCIEX QTRAP® 6500 System and the AB Sciex TripleTOF TM 5600 System. NORTH AMERICAN INDUSTRY CLASSIFICATION SYSTEM (NAICS) CODE: The intended procurement is classified under NAICS Code: 334516, with a Size Standard: 500 Employees. REGULATORY AUTHORITY: The resultant contract will include all applicable provisions and clauses in effect through the Federal Acquisition Circular (FAC) 2005-68 effective June 26, 2013. This acquisition is conducted under the procedures as prescribed in the FAR Subpart 13.5-Test Program for Certain Commercial Items and the FAR Subpart 12-Acquisition of Commercial Items at an amount exceeding the simplified acquisition threshold ($150,000) and not exceeding $6.5 million. STATUTORY AUTHORITY: This acquisition is conducted under the authority of 41 U.S.C. 253(c)(1) under provisions of the statutory authority of the FAR Subpart 6.302-1 Only one responsible source and no other supplies or services will satisfy agency requirements. DESCRIPTION OF REQUIREMENT: AB SCIEX QTRAP® 6500 System: • Full triple quadrupole mass spectrometer functionality including multiple reaction monitoring (MRM), neutral loss and precursor ion scanning. MRM experiments will be used for detecting and quantitating drugs, drug metabolites and other analytes of interest at low to sub femtogram levels • Full linear acceleration ion trap functionality, including MS/MS (enhanced product ion scanning or EPI) and MS3. This functionality is critical for streamlining the assay development process and for acquiring confirmatory scans that can be matched to entries in drug databases. A library match can be key to defending data in court. • Triple-Trap scanning is unique to the QTRAP technology and will be utilized to provide higher quality data in a shorter amount of time than other mass spectrometers. In this mode, MRM scans are used to detect or quantitate the presence of specific analytes. Once detected, the instrument switches to ion trap mode to perform a high sensitivity MS/MS (EPI) experiment, thus confirming the structure of the compound. Enhanced resolution and MS3 scans can be incorporated into this workflow. A particularly powerful and unique aspect of this workflow is the similar detection limits that can be reached for the EPI and MRM scans, allowing for confirmatory EPI scans to be acquired on virtually any analyte that is detected by MRM. In Triple-Trap mode the 6500 QTRAP is able to scan 100 MRM transitions followed by a precursor scan (500 Da) and a neutral loss scan (500 Da) followed by two Enhanced Product Ion scans in 1.5 seconds. This workflow is unique to QTRAP technology and will provide more comprehensive data sets for casework than any other mass spectrometer. Another application of triple trap scanning will be for the development of new assays that detect new or more specific drugs or drug metabolites. Precursor ion or neutral loss scanning experiments can be used to discover new "candidate" analytes and trap scanning can be used for structural confirmation of the candidates. • Quantitation by MRM3 is only available on QTRAP instruments. MRM3 scans can be applied to complex assays that are subject to interferences from co-eluting compounds or matrix ions. Standard triple quadrupole mass spectrometers do not have MRM3 functionality so are more prone to interferences and therefore are more likely to produce inaccurate or erroneous results. • Enhanced Resolution Scanning: using the linear ion trap the QTRAP can acquire spectra with resolving power (measured FWHM) >9000 in 10s of milliseconds. This functionality will be important during method development for detecting and eliminating interferences and for chemical structure confirmation. No triple quadrupole mass spectrometer has this capability. AB Sciex TripleTOF TM 5600 System: The 5600+ Triple TOF is a high sensitivity, fast scanning time of flight mass spectrometer which utilizes several of the same technologies as the 6500QTRAP. The patented TurboV ion source and LINAC Collision Cell are common to both instruments. The 5600+ also utilizes the unique Accelerator TOF Analyzer which benefits from high acceleration voltage and a 40GHz multichannel TDC detector. Background: The Government Requiring Agency needs an AB SCIEX QTRAP® 6500 System and an AB Sciex TripleTOF TM 5600 System for vital laboratory research. Specifically, the Government Requiring Agency needs the items to support analysis of hundreds of diverse biological matrices collected in our clinical research endeavors. Purpose and Objectives: The Government Requiring Agency needs two liquid chromatography tandem mass spectrometers to support analysis of hundreds of diverse biological matrices collected in our clinical research endeavors. One instrument must be a high-resolution tandem mass spectrometer and the other instrument must be a quadrupole linear ion trap design. These instruments will be used for quantification and qualification studies investigating pharmacokinetics, correlating drug concentrations to observed drug effects, identifying new biomarkers for documenting drug intake and characterizing drug metabolism with both clinical and in vitro hepatocyte samples. Period of Performance/Delivery and Contract Type: The delivery will occur within 60 days. The contract type is Firm-Fixed-Price. Project Description: The Qtrap 6500 will be applied for quantification of drugs and metabolites via triple quadrupole multiple reaction monitoring and for structural elucidation of unknown, novel metabolites via ion trap capabilities. The Government Requiring Agency pharmacokinetic studies require sensitive and specific simultaneous quantification of drugs and metabolites in specimens collected during IRB-approved clinical studies. Drug concentrations will be correlated to observed subjective and physiologic drug effects. The instrument must be able to do rapid analysis of many different analytes- for instance 54 synthetic cannabinoids and metabolites in urine. The Government Requiring Agency conducts investigations in diverse matrices including whole blood, oral fluid, plasma, meconium, placenta, brain and other matrices to determine windows of drug detection and to assess time of last drug intake. Finally, the Government Requiring Agency will use this instrument to identify unknown, novel drug metabolites to discover new biomarkers for identifying drug exposure. The TripleTOF 5600+ will be applied for both quantification of known drugs and metabolites via MS/MS and for structural elucidation of unknown, novel metabolites via high resolution TOF capabilities. The Government Requiring Agency pharmacokinetic studies require sensitive and specific simultaneous quantification of drugs and metabolites in specimens collected during IRB-approved clinical studies. Drug concentrations will be correlated to observed subjective and physiologic drug effects. The Government Requiring Agency conducts investigations in diverse matrices including whole blood, urine, oral fluid, plasma, meconium, placenta, brain and other matrices to determine windows of drug detection and to assess time of last drug intake. Most importantly, we will use this instrument to identify unknown, novel drug metabolites to discover new biomarkers for identifying drug exposure and for emerging drugs (synthetic cannabinoids (SPICE) and cathinones) high-throughput screening. Other Important Considerations: Not Applicable. CONTRACTING WITHOUT PROVIDING FOR FULL AND OPEN COMPETITION: The determination by the Government to award a contract without providing for full and open competition is based upon the market research conducted as prescribed in FAR Part 10-Market Research, specifically the review and evaluation of the responses to the sources sought notice published in the FedBizOpps. The equipment can perform functions, which cannot be found in any other commercially available equipment, as follows: 6500 QTRAP: 1. Triple quadrupole instruments manufactured by other vendors are all dedicated triple quadrupole mass spectrometers and therefore are functionally limited to traditional triple quadrupole experiments. No other instrument has hybrid ion trap functionality in addition to triple quadrupole functionality and therefore is unable to perform MS3, MRM3, product ion scans at the same LOQs as MRM experiments and Triple-Trap scanning as provided by AB Sciex's state of the art instruments. All of this functionality could be used by the laboratory for research and casework, especially MRM3 and Triple-Trap scanning. Triple-Trap scanning could streamline the development of new assays and will be utilized in casework for simultaneously quantifying drugs while acquiring drug fingerprint mass spectra that can be matched to database entries. 2. The 6500 QTRAP Mass Spectrometer is covered by at least the following AB SCIEX Patents: QTRAP Related Patents: 6,177,668; 6,627,876; 6,909,089 and 7,227,130. Ion Drive Technology Related Patents: 5,412,208 ; 6,759,650 ; 7,256,395; 7,259, 371 and 8,374,799. TurboV Ion Source Related Patents: 5,412,208 and 6,759,650. LINAC Collision Cell Related Patents: 5,847,386; 6,111,250 and 7,199,361. 5600+ Triple TOF: 1. The 5600+ Triple TOF is a high sensitivity, fast scanning time of flight mass spectrometer which utilizes several of the same technologies as the 6500QTRAP. The patented TurboV ion source and LINAC Collision Cell are common to both instruments. The 5600+ also utilizes the unique Accelerator TOF Analyzer which benefits from high acceleration voltage and a 40GHz multichannel TDC detector. These proprietary features result in performance which is unmatched by any other vendor: 2. That the AB Sciex TripleTOF TM 5600 system is unique and exclusive in the mass spectrometry market space can be seen from the following unique characteristics and related patents: - The AB Sciex TripleTOF TM 5600 system has a proprietary Real Time Multiple Mass defect triggered information dependent acquisition that can selectively capture the ion having the defined mass defect, and fragment the captured ion to generate the product ion spectra (for example, US patent 7,589,318) - The TripleTOF TM 5600 System has a proprietary feature for obtaining a mass spectrum enriched with fragments while retaining the parent ion using broad fragmentation coverage in mass spectrometer using varying collision energy from low to high energies.(for example, US patents 7,199,361 and 7,351,957. - The TripleTOF TM 5600 System has another proprietary feature for obtaining real time specific identification of reaction pairs associated by specific neutral differences (for example, US patent 7, 417,223) - The TripleTOF TM 5600 System has a unique automated tool to carry out on the fly dynamic background signal exclusion in mass spectrometry data dependent acquisition (for example, US patent 7,351,956) - The TripleTOF TM 5600 System also features High pressure LINAC collision cell (for example, US patents 5,847,386 and 6,111,250) for increased speed of analysis while performing high efficiency fragmentation and eliminating cross talk. - AB Sciex has a unique MS/MSALL functionality to enable SWATH TM Acquisition (multiple US Patents Filed) which utilizes the speed of acquisition of MS and MS/MS data of the AB SCIEX TripleTOF TM 5600 System in a breakthrough for proteomics mass spectrometry users to conduct comprehensive quantitation on the entire proteome - There are number of other unique patents related to TripleTOF TM 5600 and to add a few more to the list, for example, US patents 7,256,395 and 7,391,015 3. All the above features mentioned are unique and proprietary to AB Sciex No other vendor offers these unique combination of technologies (TurboV Source, LINAC Collision Cell, Accelerator TOF Analyzer). The combination of patented features of the 5600+ Triple TOF forms an instrument which has unmatched speed and sensitivity. This is critical when analyzing drugs and their metabolites. CLOSING STATEMENT: This synopsis is not a request for competitive proposals. However, interested parties may identify their interest and capability to respond to this notice. Responses to this notice shall contain sufficient information to establish the interested parties' bona-fide capabilities for fulfilling the requirement and include: unit price, list price, shipping and handling costs, the delivery period after contract award, the prompt payment discount terms, the F.O.B. Point (Destination or Origin), the Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size. All offerors must have an active registration in the System for Award Management (SAM) www.sam.gov." A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government.
