Description: The National Cancer Institute (NCI), (CCR), plans to procure on a sole source basis with Metabolon Inc., 617 Davis Dr. Ste 400, Durham, NC 27713 to assess Cross-talk between metabolism and nutritional agents in carcinogenesis. This acquisition will be processed in accordance with simplified acquisition procedures as stated in FAR Part 13.106-1(b)(1) and is exempt from the requirements of FAR Part 6. The North American Industry Classification System code is 541380 and the business size standard is $14.0 million. Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. Period of Performance: Performance shall be for two (2) months from date of award. It has been determined there are no opportunities to acquire green products or services for this procurement. Background: NCI study titled "Cross-talk between metabolism and nutritional agents in carcinogenesis" has preliminarily demonstrated that absence of thrombospondin-1 increases tumor multiplicity and lowers survival in a murine model of colon carcinogenesis. The aggressiveness of the tumors seems to be regulated in part by difference in diet exposure. The NCI's preliminary data suggest that cell metabolism is globally affected between these phenotypes and a more in depth analysis is needed to pin point specific altered pathways. NCI has also included in the study the CD47 strain to continue in vivo studies on the differences NCI observes in mice after exposure to ionizing radiation. Project objectives: 1. Determine metabolic changes that predispose to increased tumor multiplicity and burden associated with the absence of thrombospondin-1 in the C57BL/6J-ApcMin/J (Apc-Min) strain which is highly susceptible to spontaneous intestinal adenoma formation. Also NCI plans to gain insight into the pathways that are involved in the increased survival CD47 null mice exposed to ionizing radiation. 2. A detailed and global understanding of the metabolic state of cells exposed to ionizing radiation including the parallel measurement of more than 100 biochemical analytes per sample. NCI expect a detailed analysis of cell metabolism changes as a result of radiation and the presence or absence of CD47.This study should also provide new drug targets for radiation protection therapy and colon cancer. Detailed technical requirements (Contractor Requirements). The Contractor shall provide a comprehensive analysis biochemical analytes for each submitted samples according to the standard mView. This includes: Data spreadsheet, per sample: Named biochemical detected KEGG/HMDB links Relative quantitation Heat map and statistical results for each group One page summary highlighting biochemical differences A.To achieve the contractorshall use three platform to deliver metabolite analysis: •LC-MS/MS (+ESI) •LC-MS/MS (- ESI) •GC-MS B.The tables below show the number of samples to be analyzed per study: Table I. Analysis of Mouse Lungs for Ionizing radiation study: Samples Group 1WT7 2WT+IR 8 3CD47 -/-7 4CD47-/- + IR8 For the in vivo radiation studies a total of 30 samples will be analyzed Table II. Analysis of Liver, Small Instestine, Large Intestine: SamplesGroups# of Samples per tissue 1WT High Fat8 2WT low Fat8 3TSP -/- High Fat8 4TSP -/- Low Fat8 5APC min High Fat8 6APC min Low Fat8 7TSP1-/-APC min High Fat8 8TSP1-/-APC min Low Fat8 A total of 64 samples will be analyzed per tissue for a total of 192 samples Scope of work: NCI will provide tissue for analysis. Contractor shall provide all materials and equipment for sample preparation, data acquisition, and data analysis. Reason for sole source: The requested testing and evaluation services may only be obtained directly from Metabolon Inc., which is the only uniquely qualified entity to provide global, chemocentric metabolomics service using its proprietary database to known biochemical compounds that allow for the identification of biochemical's used to understand both affected and unaffected biochemical pathways. The NCI has used Metabolon for initial in vitro studies, therefore for continuity purposes, soliciting the services from this company allows for comparing metabolite changes observed in previous data. Obtaining data from a different metabolite platform may result in inconsistent infromation. To conduct this project with another contractor would result in delays to the work already in process and duplication of costs already incurred. This notice is not a request for competitive quotation. However, if any interested party, especially small businesses, believes it can meet the above requirement, it may submit a capability statement, proposal, or quotation, which shall be considered by the agency. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform the requirement. Responses must be received in the contracting office by 10:00 AM EST on September 6, 2013. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government.
