- This notice is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6, as supplemented with additional information in the notice. This announcement constitutes the only solicitation; quotes are being requested and a written solicitation will not be issued. The procurement is being issued as a request for quotation. Submit offers on Solicitation Number: PR69367. The solicitation is being issued as a request for quotation. The solicitation documents and incorporated provisions and clauses are those in effect through FAC 2005-72 effective January 30, 2014. This acquisition will be processed under Simplified Acquisition Procedures (SAP), Acquisitions of Commercial Items and is not a Small Business Set-Aside. The North American Classification System (NAICS) code for this procurement is 541380, and the small business size is eleven million dollars. The Center for Disease Control and Prevention has a requirement for PCR technology which replaces time-consuming, subjective in-house testing with consistency and certificates of analysis under pending GMP regulation. This fulfills the cell culture and hybridoma needs of DSR and growing list of projects developed from CDC SMEs. To meet the rigorous standards of high sensitivity PCR detection with our cell samples there should be qualifications for the PCR test for each cell line tested. Because unknown factors within each cell line may contribute to false positives and/or false negatives a series of controls should be included in the testing. These should include: 1. Spike Recovery Control where An aliquot of sample is spiked with a known concentration of mycoplasma-like material to determine the efficiency of isolating mycoplasma from a given matrix. Once a defined matrix is qualified in this way, subsequent spike recovery tests can be avoided unless changes occur within the matrix composition (e.g. media components, cell density, etc.). Recommended for every sample type. 2. Positive Extraction Control where Mycoplasma-like material is isolated on its own for any given extraction process and treated in the same way as the sample. Demonstration that the extraction procedure has been done in a way that would allow for the extraction of the target DNA and that the reagents are working properly. This is necessary to ensure against false negative results. Recommended for every extraction process. 3. Negative Extraction Control where Extraction reagents, in the absence of target DNA, are carried through each step of the extraction process. This is necessary to ensure that neither the reagents nor the process are introducing any target DNA that may result in a false positive. Recommended for every extraction process. 4. Inhibition Controls A second set of PCR reactions containing DNA isolated from the client sample and spike recovery control are spiked with mycoplasma-like material. This is done to ensure the absence of inhibitory substances carried through the isolation that might delay or completely mask a positive signal. Recommended for every sample and spike recovery control. 5. Positive PCR Control where A PCR reaction that is run on each plate to confirm the functionality of the PCR reagents and detection system. A measure taken to minimize the possibility of false negatives. For instances in which the positive extraction control did not perform optimally, this control can limit the problem to the isolation procedure. Necessary for every run. 6. Negative PCR Control where a PCR reaction that is run on each plate to confirm that the PCR reagents have not introduced target DNA or caused high levels of background. A measure taken to minimize the possibility of false positives. For instances in which the negative extraction control is positive, this control will isolate the problem to the isolation process. Necessary for every run. CLIN 0001: 8 tests per submission for a total of 96 tests per year.
