INTRODUCTION: THIS IS A PRE-SOLICITATION NON-COMPETITIVE (NOTICE OF INTENT) SYNOPSIS TO AWARD A CONTRACT WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME). THIS IS A NOTICE OF INTENT, NOT A REQUEST FOR A PROPOSAL. A SOLICITATION DOCUMENT WILL NOT BE ISSUED AND PROPOSALS WILL NOT BE REQUESTED. The National Heart, Lung, and Blood Institute (NHLBI) Office of Acquisition (OA) on behalf of the National Human Genome Research Institute, (NHGRI), intends to negotiate and award a purchase order on a noncompetitive sole source basis to Pacific Biosciences, 1380 Willow Road, Menlo Park, California 94025. (BRAND-NAME: PACIFIC BIOSIENCES, PACBIO RSII SEQUENCER SYSTEM) Procurement: 1. PacBio RSII System including instrument, Blade Center, PacBio Software Suite and Installation Kit with 12-Month Standard Service Contract, Part Number: 100-210-100, Quantity: One (1) Each. 2. Trade-In Genome Sequencer FLX 454, NIH Property Form 1872: DOC Number: 2014-178. 3. UPS, 200/208-240 VAC, 1 Phase 60 HZ, -PB Instrument Grade UPS, Part Number: 100-217-000, Quantity: One (1) Each. 4. On-Site System Training (3 Days), Part Number: 100-125-100, Quantity: One (1) Each. PERIOD OF PERFORMANCE : Delivery within 4-6 weeks upon award BACKGROUND: The National Institute of Health (NIH) mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability. The National Human Genome Research Institute (NHGRI) mission has expanded to encompass a broad range of studies aimed at understanding the structure and function of the human genome and its role in health and disease. NHGRI supports the development of resources and technology that will accelerate genome research and its application to human health. A critical part of the NHGRI mission continues to be the study of the ethical, legal and social implications (ELSI) of genome research. NHGRI also supports the training of investigators and the dissemination of genome information to the public and to health professionals. The National Human Genome Research Institute NHGRI), NIH Intramural Sequencing Center (NISC) is a high-throughput sequencing center supporting all institutes within the NIH. NISC has served the NIH community for over 15 years as a source for high-quality large-scale sequencing and analysis. As next generation sequencing applications expand, so does the demand for NISC services. NISC currently has one (1) HiSeq 25000, three (3) MiSeq Systems, and four (4) HiSeq 2000 Systems which are proven platforms for genetic analysis and functional genomics. However these instruments provide a maximum read length of 300 bases. Complex genomes cannot be de novo assembled into one piece with reads of this length. Typically even small microbial genomes are in 50-300 pieces even with 100x coverage. Also there are ALWAYS missing pieces to the genome. Pacific Biosciences Pac Bio RS II Systems is a Single Molecule, Real-Time (SMRT®) DNA Sequencing System that provides the highest consensus accuracy and longest read lengths of any available sequencing technology. The instrument features high performance optics, automated liquid handling, and an environmental control center, all directed through an intuitive touchscreen interface. The computational brain responsible for primary data analysis, called the Blade Center, is also included. This allows for seamless integration of performance enhancements through chemistry and software advances. The instrument contains three primary user access points: • RS Touch uses a touchscreen interface to start and monitor runs at the instrument. • Reagent and Sample Drawer for plate loading. • SMRT Cell and Tip Drawer for consumables loading. The PacBio RS II produces read lengths in excess of 10,000 bases. The sample preparation for sequencing does not require amplification, so there is no risk of artifact introduction. The PacBio RS II sequencing technology has the added advantage of identifying modified bases which among other things can indicate the activity of genes in the tissue from which the DNA was derived. The accompanying computer provides real-time processing to reduce the data to a manageable size for downstream analysis. PROJECT DESCRIPTION: Pacific Biosciences produced 10 fully assembled genomes in just 2 weeks. The data lead to the additional resolution of multiple plasmids present in each isolate, which is critical for understanding modes of antibiotic resistance transmission across strains. In addition, investigators in NIAID have had impressive success in sequencing the full HIV sequence spectrum in patients. These accomplishments can only be achieved with read lengths well over 1000 bases The PacBio RS II allows researchers to obtain extremely long sequence reads that open up many new possibilities in the field of genomics. The simple library method involves no amplification and can be performed in 1day in most cases. These features allow NISC to support microbial genome, virus genome, RNA isoform, methylation studies and more that have never been possible before. Contractor Requirements: • The Contractor shall provide uncrating and set up of the instrument. • The Contractor shall validate that the instrument is operating fully and will provide full training for library construction, instrument operation, and bioinformatics post-run processing and assembly. Inspection and Acceptance Requirements: • The Contractor shall deliver instrument, computer and accessories in good working condition. The Government will verify satisfactory performance of the instrument. REGULATORY AUTHORITY : This acquisition is conducted under the authority of 41 U.S.C. 253(c) as set forth in FAR Part 6.302-1, only one responsible source and no other supplies or services will satisfy agency requirements. ADDITIONAL INFORMATION: Industry Classification (NAICS) Code is 334516, Analytical Laboratory Instrument Manufacturing and the Small Business Size Standard is 500 Employees. The acquisition is being conducted under FAR Part 13, simplified acquisition procedures, therefore the requirements of FAR Part 6 B Competitive Requirements are not applicable (FAR Part 6.001). The resultant Contract will include all applicable provisions and clauses in effect through the Federal Acquisition Circular (FAC) 05-73 (April 29, 2014). This synopsis is not a request for competitive proposals. However, interested parties may identify their interest and capability to respond to this notice. Responses to this notice shall contain sufficient information to establish the interested parties' bona-fide capabilities for fulfilling the requirement and include: unit price, list price, shipping and handling costs, the delivery period after contract award, the prompt payment discount terms, the F.O.B. Point (Destination or Origin), the Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size. All offerors must have an active registration in the System for Award Management (SAM) www.sam.gov." A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. The information received will normally be considered solely for the purposes of determining whether to proceed on a non-competitive basis or to conduct a competitive procurement.
