- National Institutes of Health is seeking to obtain information regarding: (1) the availability and capability of qualified small business sources; (2) whether they are small businesses; HUBZone small businesses; service-disabled, veteran-owned small businesses; 8(a) small businesses; veteran-owned small businesses; woman-owned small businesses; or small disadvantaged businesses; and (3) their size classification relative to the North American Industry Classification System (NAICS) code of 334516 with a size standard of 500 employees for the proposed acquisition of a MiSeq Reagent kit v3 (600 cycle) including paired-end reagent plate. Your responses to the information requested will assist the Government in determining the appropriate acquisition method, including whether a set-aside is possible. An organization that is not considered a small business under the applicable NAICS code should not submit a response to this notice. Background: The mission of the Cellullar Biology and Viral Immunology Sections is to gain fundamental insight into: Cell biology: by studying MHC class I associated antigen processing pathways, and the folding, trafficking, and assembly of viral proteins, particularly IAV (IAV) hemagglutinin (HA). Cellular immunology: by studying the activation, repertoire, and function of anti-viral T cells and B cells responses as well as Influenza Virus Biology: by studying IAV antigenic variation and pathogenicity Laboratory based projects/clinical support: Identify vaccination modalities that generate heterosubtypic immunity in guinea pigs. Assess capacity of heterosubtypic immunity to limit IAV viral replication and transmission in GPs. Assess effects of BNAbs on IAV evolution and escape. Contractor Requirements • 22-25 million reads per run (e.g. clusters passing filter) • 13.2-15 Gb data per 2 × 300 bp throughput per run • single-read or paired-end runs • imaging from top, bottom, or both surfaces of flow cell original, micro or nano flow cells can be used to select image area utilization and thus select data output levels • Read-length is fully adjustable from 36-300 base pairs Overlapping 2 × 300 bp reads provides ability to generate up to 600 bp reads on the same fragment • Uses reversible terminators and a highly efficient DNA polymerase • The DNA polymerase is modified for efficient addition of nucleotides with cleavable fluorescent dyes and reversible terminators • Sequencing reactions are performed in a self-contained single channel flow cell environment • Fluorescent dyes on the nucleotides are cleaved after imaging • Reversible terminators are removed to allow chain extension • Competitive addition from a pool of all four reversible terminator nucleotides labeled with four different fluorescent dyes • Sequenced DNA templates are copied to generate complementary strands, enabling paired-end sequencing • Forward DNA strands are selectively washed out of the flow cell Accurate variant detection enabled by Sequencing by Synthesis (SBS) chemistry • Competitive nucleotide addition with a reversible terminator technology Exceptionally high quality score distributions at every read length • > 90% of bases with Q scores > 30 (1 × 36 bp) • > 90% of bases with Q scores > 30 (2 × 25 bp) • > 85% of bases with Q scores > 30 (2 × 100 bp) • > 80% of bases with Q scores > 30 (2 × 150 bp) • > 75% of bases with Q scores > 30 (2 × 250 bp) • > 70% of bases with Q scores > 30 (2 × 300 bp) Any potential offers should respond with their capability statement by June 30, 2014.
