- Note: This is a repeat advertisement of N02RC42546-83 posted on June 18, 2014 The National Cancer Institute (NCI), Center for Cancer Research (CCR), Laboratory of Cellular and Molecular Biology (LCMB), plans to procure an integrated upgrade package for a Government owned Nikon Ti Eclipse TIRF microscope on a sole source basis with Nikon Instruments Inc. 1300 Walt Whitman Road - Melville, NY 11747. This acquisition will be processed in accordance with commercial items prepared in accordance with the format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; quotes are being requested and a written solicitation will not be issued. The pre-solicitation number is N02RC42580-83. This solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular (FAC 20) 05-72, (January 2014) Simplified Procedures for a Commercial Item. The Government contemplates only one award of a Firm-Fixed Price contract. The Associated North America Industry Classification Code (NAICS) 334516 and the Business Size Standard are 500. It has been determined there are no opportunities to acquire green products or services for this procurement. Delivery will be 60-90 days from date of award. The Laboratory of Cellular and Molecular Biology (LCMB) investigates normal cellular signal transduction and aberrations in signaling pathways that are associated with neoplasias in humans and model systems. LCMB scientists have been very successful in using imaging approaches to study these topics. Five years ago, NCI acquired a Nikon TIRF microscope and the LCMB began to develop techniques using Photo Activation Localization Microscopy (PALM), which allows localization of genetically encoded proteins to within 20 nm. This project has been very successful, resulting in several publications. As a result, NCI would now like to expand its super-resolution capabilities to include Stochastic Optical Reconstruction Microscopy (STORM), which allows localization of antibody labeled molecules. STORM will allow NCI to resolve the location of endogenous proteins and phosphorylated proteins to within 20 nm, a significant expansion of the LCMB high resolution capabilities. Projects are underway to determine the composition and time dependent changes in protein complexes formed during the activation of receptors at the single molecule level. Once the upgrade is implemented, the system can be used for both PALM and STORM imaging by any investigator who uses the LCMB Microscopy Core. The Laboratory of Cellular and Molecular Biology (LCMB) requires improvement to the LCMB's TIRF microscope for experiments involving the observation of endogenous or phosphorylated proteins with resolution of structures to within 20-40 nm in three dimensions by Stochastic Optical Reconstruction Microscopy (STORM). These experiments require imaging equipment with sufficient sensitivity to visualize single molecules, lasers powerful enough to use the STORM technique and software capable of producing STORM images. The integrated upgrade package from Nikon Instruments Inc. possesses the following salient characteristics: 1) Modifications must allow use of current microscope (Nikon Eclispe Ti), 100X TIRF objective NIH decal #01751173. 2) The combined outcome of these modifications must be to allow both PALM and STORM imaging with 3D capabilities, while maintaining TIRF and epifluorescence capabilities, 3) Addition of STORM components: Ti-TIRF motorized illuminator Unit, TI-N-STORM kit, iXON DU897 Ultra 512x512 EM-CCD camera, stage up brackets, 4) Addition of filter wheels: Lambda 32 nm filter wheel, Lambda 25 nm emission filter wheel, TI emission adapter, HS-625 filter wheel setup for digital triggering with external breakout box and adaptors for same, excitation adapter 5) Lambda 10-3 controller, 6) TIRF dichroics and filter sets: STORM quad filter set, STORM 647 filter set, 89000 Sedat Quad ET filter set mounted TiTE2000, Custom 440/514/561/649 filter MTD TIRF, FL STIC filter set, ET430/24x, ET CFP Excitation Filter 32mm, YFP Exciter 32mm, ET480/40m, Hard Coat Emission Filter, ET525/36m Emission Filter, max blocking for 488 & 561 laser lines. 25mm diameter, max 5mm thick, ET705/72m Emission Filter, Far Red Emission for SpectraX quad band 25mm, ET500LP, ET 500nm Long Pass Emission Filter 25mm, ET520LP, Long Pass Emission Filter 25mm, ET570LP, Long Pass Emission Filter 25mm, ET545/40m, Band Pass Emission Filter 25mm, Polarizing Filter (25 mm or smaller), 7) Piezo Z for 3D STORM: Piezo Stage Insert with USB Controller, Petri Dish Holder 35MM for Piezo Stage, Slide Holder 1x3 for Piezo Stage, Cover Slip Holder 1x1 for Slide Holder 1x3 for Piezo Stage, Multi-Well Plate Holder for Piezo Stage, 8) 6 Line solid state laser combiner: LUNB Solid state laser combiner: 405S/445S/488H/514S/561H/647X A/A/F, NIDAQ LUnB Connection kit: includes breakout box for NI6723 and NI6353cards, all connectors, and interlock cable, NI PCIe-6353 DAQ card - 32AI and 48 I/O lines. This is not a solicitation for competitive quotations. However, if any interested party believes they can meet the above requirement, especially small businesses, they may submit a tailored statement of capabilities. All information furnished must be in writing and must contain sufficient detail to allow the NCI to determine if it can meet the above unique specifications described herein. An original and one copy of the capability statement must be received in the NCI Office of Acquisitions on or before 11:00 AM EST on, August 8, 2014.
