- The National Marine Fisheries Service (NMFS) intends to issue a sole source award utilizing Simplified Acquisition Procedures to purchase from the University of Arizona. The purchase will be to to genotype up to 2,400 Chinook salmon DNA samples for 43 SNP markers using the two predesigned MALDI-TOF genotyping panels. The scope of the work is to genotype up to 2,400 Chinook salmon DNA samples for 43 SNP markers using the two predesigned MALDI-TOF genotyping panels. Primer sequences will be provided for the MALDI-TOF SNP assays and the contractor is responsible for ordering necessary primer stocks. If required (never been done before), as proof of principle, the contractor will test the two panels (all 43 assays) by genotyping 190 known Chinook DNA samples (provided by the Point of Contact (POC)). DNA quantitation of all samples will be performed by the contractor. After the initial test, the contractor will furnish the scored genotypes and cluster with the known genotypes. The contractor should be willing to work with the POC to make scoring changes based on the cluster plots. To continue this project, the POC must approve the quality of the genotypes. Without extenuating circumstances, all 43 assays in the 2 MALDI-TOF panels should score in at least 95% of the fish with greater than 99% accuracy. After acceptance of the assays, DNA from up to 2,800 additional Chinook salmon will be provided to the contractor by the POC. All DNA quantitation should be performed by the contractor and results supplied to the POC. All analyses should include at least 10 DNA controls with known genotypes (provided by the POC). For quality control, all 43 assays should score in at least 90% of all the analyzed fish (dependent on DNA quality) and the controls should score with greater than 99% accuracy. University of Arizona is the only vendor capable of providing these services based one the fact that The University of Arizona Genetics Core has developed specific MALDI-TOF assays to ascertain the 43 single nucleotide polymorphisms represented in the Chinook salmon genetic baseline. They are a not-for-profit academic core facility designed to process and produce data for which the National Marine Fisheries Service retains sole ownership. Through our market research, they are the only facility in the country that has the necessary experience in MALDI-TOF genotyping Chinook or Pacific salmon for those genetic markers. This award will be for a base period of 13 plates with an option 1 for an additional 10 plates and a second option for an additional 9 plates. Exercising of options are dependent on number of samples collected at sea. The government acknowledges a possibility of awarding option 1 and/or 2 with initial award as additional samples may arrive during the solicitation period. This notice of intent is for information purposes only, therefore, no solicitation will result from this announcement. All sources eligible to provide this service must respond in writing, response must be supported by clear and convincing evidence to clearly be able to provide services outlined above. A request for documentation will not be considered an affirmative response. If no responses are received, within 15 days after publication of this notice, to the effect that comparable services are available, and that it is more advantageous to the Government than obtaining these services through a sole-source contract, then a sole source acquisition award will be made. This is a simplified acquisition with an estimated value of less than $150,000.00 total base and options.
