PART 1 - Intro The Food and Drug Administration (FDA) has a requirement to purchase one (1) mobile fully integrated and automated Disposable Bioreactor System with an Alternating Tangential Flow (ATF) Filtration system for fed batch or continuous perfusion culture operations of mammalian cell cultures. This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6, (STREAMLINED PROCEDURES FOR EVALUATION AND SOLICITATION FOR COMMERCIAL ITEMS) as supplemented with additional information included in this notice. This announcement constitutes the only solicitation. This solicitation is issued as a Request for Quote (RFQ), using the Simplified Acquisitions Procedures of FAR 13. The solicitations document/incorporated provisions and clauses are those in effect through the current Federal Acquisition Circular: FAC 2005-76. The associated North American Industry Classification System (NAICS) Code is 325414, Biological Product (Except Diagnostic) Manufacturing with a Small Business Size Standard is 500 employees. This acquisition is set aside 100% for small business concerns. The vendor shall provide sufficient consumables to support 12 runs during the base year's period of performance. The bioreactor vessel suitable sterilization consumables that are pre sterilized and ready for use. For reusable probes and accessories, external sterizization with an autoclave, sterilization pouches and connectors shall be provided to allow an aseptic installation outside of a biological safety cabinet in a laboratory with standard room air. o Capable of operating volumes between 10L and 15L, with a load level sensor integrated with a media feed pump to maintain adequate volume levels throughout the process. o The system shall be capable of operating with suspension and adherent mammalian cell lines, and able to operate in either batch, fed batch, or perfusion (continuous) modes. The bioreactor vessel should be of disposable material to avoid the need for cleaning and autoclaving. o Standard pH, Dissolved Oxygen, Temperature monitoring and algorithms for control. Condensing or heating of exhaust filters should prevent clogging and over pressurization throughout the cycle. o Due to shear sensitivity of cell lines utilized, the system should not utilize traditional stirred tank agitation, but an ultralow sheer agitation approach to prevent agitation damage to the cell culture. o A capacitance/conductivity system shall be provided and integrated to allow real time online monitoring of viable and non-viable cells within the reactor system. This system shall be used as a trigger for making perfusion based decisions in both an automated or manual fashion. o Feeding Pumps: At least 3, with adjustable speeds, and individual feeding ports for base control, media feeds, and supplementation. The feed pumps should be programmable and integrated with the system to receive automated commands to turn on, off, or change rate. o Sampling Pump: A sampling pump (separate from the feeding pumps above) and retention system shall be integrated into the system to allow operator directed or automated sampling procedures. The configuration should also be compatible with external auto samplers as well to allow additional automated sample delivery to an external nutrient analyzer currently owned by the division (NOVA FLEX Nutrient Analyzer). o Ability to monitor and automatically implement (with a manual option) the removal of living and dead cells when densities surpass target ranges to maintain culture health. The removed material should separate cells from harvested culture fluid without returning cells to the system. The system must also minimize product loss when removing cells and provide a method to suitably clarify the separated harvested culture fluid or aseptically return the separated fluid to the bioreactor. The clarified material will then also be available to the downstream FPLC purifier for automated loading and purification to reduce product loss without additional intermediate steps. o At least 1 harvest line should be integrated directly into the ATF system. The rate of perfusion from the bioreactor to the ATF must be controlled through automated integration of an appropriate pump, valve, or equivalent (with a manual option) into the system to ensure the desired flow rates are maintained throughout the culture. • Alternating Tangential Flow (ATF) System to clarify harvested cell culture fluid o The ATF connection to the bioreactor harvest line must be aseptic to prevent contamination, and adequate to handle necessary flow rates of a perfusion culture of up to 30L per day with minimal dead volume or pressurization. o The ATF holofiber component must be capable of preventing living and dead cells from leaving the bioreactor and moving down stream to the purifier. o The ATF should have standard control systems to minimize shear effects on cells, allow diaphragm rate adjustment, and other necessary changes throughout the process. o Material exiting the system should be clarified sufficiently to enter a sterile holding tank or equivalent. The tank will act as a physical link to supply clarified material between the vendor supplied system and further downstream purification utilizing a standard Fast Protein Liquid Chromatography (FPLC) purifier (supplied by the FDA). The final holding tank or equivalent is expected to be sterilizable, robust throughout the process, with sufficient entries, exits, and exhaust to prevent over pressurization, for aseptic manipulations and connectors. o Mobile cabinet with casters that shall house the unit. Maximum Dimensions of Mobile Cabinet: 48" long x 24" deep x 72" tall. • Peripherals, Accessories, and Consumables: o The vendor shall provide compatible peripherals necessary to ensure an integrated turn-key system. o This includes but is not limited to: peristaltic pumps, physical connectors, balances, tanks, intermediates, controllers and control systems. o The vendor shall provide adequate and appropriate consumables to ensure installation, training, and initial operation of the integrated system. In addition, 12 disposable bags and 12 cartridges shall be included. o All software and licenses required to integrate, operate, analyze and export the results shall be provided. • Shipping o Crating and delivery shall be included in this proposal to the delivery address. INSTALLATION: The vendor shall provide inside delivery, installation, setup, and optimization of the system. Warranty period will commence after the installation of the system is accepted by the Program.  Crating and shipping to the delivery address  On-site installation and integration of hardware and software.  Integration: o Hardware- All physical connections must be made and verified by the vendor to ensure physical operability and protection from environmental and microbial contamination. o Software/Data Transfer- Ole for Process Control (OPC) or a compatible approach shall be used to integrate third party software components to allow data exchange across platforms to feed automated control algorithms. This integration should also include the division's existing NOVA Flex (which uses an OPC platform) into the system to support nutrient feeding algorithms. Automated systems shall also provide an option for manual "over-ride" or user inputs where appropriate. o Integration shall be provided by OEM contractors with experience in bioreactor, clarification, purification, and OPC integration. o All hardware and physical connections required to be sterile must be sterilizable or disposable pre-sterilized materials to ensure an aseptic cell culture environment throughout the process.  After installation vendor is required to demonstrate full system functionality by conducting an IQ/OQ validation. TRAINING: The Contractor shall provide on-site training to approximately 5 laboratory personnel within one month after the system has been delivered, installed and accepted by the Program.  The purpose of the training is to familiarize staff with the proper use of the system including:  system operations (including software)  calibration  optimization  basic and routine preventative maintenance procedures  cleaning requirements  any safety and related safeguards as appropriate;  information on what users should do if a unit or the system is not functioning properly  any other topics deemed necessary to familiar FDA employees with the system.  This training should last 16 hours.  Contractor shall provide all training materials (web based or written) 7 days before installation. Warranty and Maintenance/Support Agreement: 1. Warranty: Vendor shall include:  Trouble shooting for issues that arise during normal operation conditions  Vendor will be responsible for parts, travel, and labor for these site visits  All components of the system shall be covered by a minimum one year warranty, with full replacement of any/all components that fail or malfunction within that timeline  System software and firmware updates required for reliability 2. Maintenance/Support Agreement: Option year maintenance agreement to cover the following:  Unlimited Phone and email support during normal business hours, (Monday-Friday, 8 AM-5PM EST)  Trouble shooting capabilities based on complete knowledge of the entire system, immediate access to certified replacement parts, and immediate access to improvements and new procedures provided by the original equipment manufacturer  Corrective maintenance, service and repairs shall include all labor, travel and parts  At least one (1) preventative maintenance visit (which shall include a calibration program in which a NIST Traceable certificate will be issued by the vendor to the Government) or more as suggested by the manufacturer  All maintenance and repair work activities shall be accomplished with certified technicians/engineers following Original Equipment Manufacturer (OEM) specifications, manuals, and service bulletins, using OEM replacement parts, components, subassemblies, etc.  System software and firmware updates required for reliability.  Service Records and Reports: The Contractor shall commensurate with the completion of each service call (inclusive of warranty service work) provide the end user of the equipment with a copy of a field service report/ticket identifying the equipment name, manufacturer, model number and serial number of the equipment being serviced/repaired and detailing the reason for the service call, a detailed description of the work performed, the test instruments or other equipment used affect the repair or otherwise perform the service, the names (s) and contact information of the technician who performed the repair/service, and for information purposes, the on-site hours expended and parts/components replaced. In addition, the Contractor shall provide a written report to the FDA COR and Contract Specialist, summarizing all maintenance and repair activities (including warranty work) each time service and/or repair is performed 3. Consumables: 12 disposable bags and 12 cartridges: Items must be sterilized and shipped ready to use. For the purpose of this PART, (COR - to be determined upon contract award) is the authorized representative of the Contracting Officer. 4.2. Inspection and acceptance will be performed at the locations identified in 6.1.3. Acceptance is communicated in writing unless otherwise indicated in writing by the Contracting Officer or COR within 30 days of receipt. 4.3. DELIVERIES Satisfactory performance of this contract shall be deemed to occur upon delivery, installation of the systems described in PART 3 of this solicitation, training and upon acceptance by the COR, of the services and reports required at the dates specified within PART 3 of this request for proposal. 5.2.
