Description
Background: This microscope will be used to visualize histology and immune-stained mouse tissue samples. In our laboratory we examine the molecular and cellular responses in inflammatory diseases such as uveitis and arthritis. Scoring histopathology of such diseases is a critical readout for many experiments and a technique we use on a daily basis. Experiments involving immune-staining allow us to fluorescently label molecules and/or cells of interest and the fluorescent microscope is critical to our being able to visualize such results. It has been determined that a computer is essential for the operation of the microscope-but this computer will not be connected to the VA Network. Several specifics regarding the chosen scope are pertinent to the type of research we do. The microscope chosen has transmitted white light for brightfield microscopy of tissue sections stained with conventional colored histology stains. It also has appropriate filters in place for viewing of fluorochromes in three different color ranges (blue, green, and red) for fluorescent tissue samples, either from animals genetically-engineered to express fluorescence or tissues that are immune-stained for specific molecular or cellular markers. Two cameras (one highly-sensitive monochromatic for low light samples plus one color) are needed to transmit images to a computer where they will be visualized, analyzed, and archived. This is also a HDMI output camera that enables high definition output for movies, thus a Digital Video Recorder is included in the quote/specs. Also, a direct coupling device is required, one which does not utilize light guidance which is degraded by light source and would require further replacement. This computer is essential for the operation of the microscope and will not be connected to the VA network. "All product offering shall fit with preexisting equipment - monochrome camera (originally purchased from Germany, Kappa, Gleichen, Germany). Updated to now include PCO-Pexelfly camera and Zeiss Axio microscope, which will allow movie capabilities as well. "All panels able to accommodate electrical service "Contractor shall ship the system and provide delivery and installation. Installation by the contractor will involve configuring microscope with camera and camera coupler(s) and coinciding objectives; installation of software. Moreover an important aspect of the installation also involves training on the multiple uses of microscope and fluorescence vs monochromatic capabilities of camera and subsequent image processing relevant to our research program that involves molecular and genetic analysis of cellular basis of eye and joint disease (i.e. uveitis and arthritis, respectively). "Contractor shall utilize environmentally preferable products made from biopreferred or biobased materials to the maximum extent possible without jeopardizing the intended end use or detracting from the overall quality delivered to the end user.
