- Description
- The Laboratory of Receptor Biology and Gene Expression was established to develop a competitive research program in the areas of chromatin structure and gene regulation, the cell biology of transcription factors, and the organization and structure of the mammalian nucleus. The laboratory supports a diverse and highly successful research program in the areas of nuclear structure, single molecule studies, chromatin structure and function, protein dynamics in living cells, structure and distribution of chromosomes, chromosome damage, higher order chromatin structure and the genome wide organization of regulatory elements. Contributions from our laboratory continue to have major impact. The QX200 Droplet Digital PCR (ddPCR TM ) System provides an absolute measure of target DNA molecules with unrivaled precision and sensitivity. The QX200 Droplet Generator partitions samples into thousands of nanoliter-sized droplets, each of which is an independent PCR reactor. After PCR on a thermal cycler, droplets are streamed in single file on the QX200 Droplet Reader to count the positive and negative reactions. The QX200 Droplet Digital PCR System delivers an absolute count of the number of target DNA sequences for applications that include copy number variation, mutation detection, and gene expression analysis. Some features of the QX200 Droplet Digital PCR System that are required for NCI scientific uses are: • The QX200 Droplet Reader is the only product on the market that can analyze 96 samples in a 96-well plate format in less than 2.5 hours. This is a required feature for us when conducting experiments. • QX200 Droplet Generator partitions a 20 μl PCR reaction into 20,000 nanoliter-sized droplets. • QX200 System can accurately count target copies when multiple target copies are distributed in a droplet. • Droplets are PCR amplified in bulk on a thermal cycler and is the only system that can process 96 samples in under 5 hours • QX200 Droplet Reader illuminates and detects fluorescence from each individual droplet. The system uses two light-emitting diodes for illumination and differentially detects emission using two filtered multipixel photon counters (one for each channel). • QX200 System has a linear dynamic range of 5 orders of magnitude for detection of input samples • QX200 System can detect one copy of target sequence from DNA samples. • One 96-well plate can be used to analyze approximately 1,500,000 droplets. • DNA from unread droplets can be recovered for downstream applications. • Bio-Rad Laboratories, Inc. is the original manufacturer and sole supplier of the QX200 Droplet Digital PCR System II.
