Description
THIS IS NOT A REQUEST FOR QUOTE: This is a special notice for a commercial item. The North American Industry Classification System (NAICS) Code for this acquisition is 334516 and the small business size standard is 500 employees or less. The USDA, Agriculture Research Service (ARS), Daniel K. Inouye, Pacific Basin Agricultural Research Center in Hilo, Hawaii intends to sole source a Bacterial Artificial Chromosome (BAC) Library to Intact Genomics, Inc. This company has been found to use a unique method to create bacterial artificial chromosomes (BAC) Libraries by preparing the target organism DNA by searing rather than by enzyme digestion. Since enzyme restriction sites are not distributed evenly across any given genome, the resulting libraries do not contain representation of the entire genome- there are missing pieces with the enzyme restriction method used by other companies. With the random shear method utilized by this company, the coverage of the genome is unbiased and more likely statistically to contain the entire genome withing the number of clones (184,320 with estimated 100 kb insert) provided by this company. The company also uses a vector for cloning that has a unique property that selects against empty vectors, ensuring a better chance of achieving each of the 184,320 clones to contain inserts. We need a genomic library that is comprehensive in order to identify and mine genomic sequences for our research. The USDA, Agriculture Research Service will be acquiring the following: 001: Creation of an unbiased (Bacterial Artificial Chromosome or BAC) genomic library of Anthurium species consisting of 184,320 clones, a 3.6 x genome coverage based on an average insert size of 100 kb. The above service includes making High Molecular Weight DNA from customer supplied plant tissue, use of proprietary methods to generate randomly sheared DNA, ligate fragments into a BAC cloning vector and transformed into E. Coli Strain DH10B cells. 002: Plate out the BAC library and array positive colones in 384 well format plates. The above service includes the tittering of each positive BAC clone into bar coded 384 well plates and stored at 80 degrees C. 003: Shipping of library on dry ice. All interested parties are encouraged to provide information on their capabilities to meet this requirement. In order to be considered a potential source, interested parties must submit clear and convincing documentation demonstrating the capabilities to satisfy the requirement specified above and advantages of the Government to consider another source. The documentation shall include prices and descriptive literature containing sufficient technical documentation to establish a bona fide capability to meet this requirement. Only written responses will be considered. All responses from responsible sources will be fully considered. As a result of analyzing responses to this synopsis, the contracting officer may determine that a solicitation will be issued. If a solicitation is issued, no additional synopsis will be published. Any such solicitation will be issued to the intended schedule vendor and all firms that respond to this synopsis. If no affirmative written response is received by Thursday, June 22, 2015, an award will be pursued with the company mentioned above.
