SOURCES SOUGHT NOTICE TEMPLATE 1.Solicitation Number:HHS-NIH-NIDA-SSSA-SS-2015-463 2.Title: AlphaLISA Albumin Assay kit, 5000 assay points 3.NAICS Code: 423460 This is a Sources Sought notice. This is NOT a solicitation for proposals, proposal abstracts, or quotations. The purpose of this notice is to obtain information regarding the availability and capability of all qualified sources to perform a potential requirement. This notice is issued to help determine the availability of qualified companies technically capable of meeting the Government requirement and to determine the method of acquisition. It is not to be construed as a commitment by the Government to issue a solicitation or ultimately award a contract. Responses will not be considered as proposals or quotes. No award will be made as a result of this notice. The Government will NOT be responsible for any costs incurred by the respondents to this notice. This notice is strictly for research and information purposes only. Background: The National Institutes of Health (NIH) is the nation's leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people's health and save lives. The National Center for Advancing Translational Sciences (NCATS), Therapeutics for Rare and Neglected Diseases (TRND) section is requesting the AlphaLISA Albumin Assay kit, 5000 assay points from PerkinElmer for the use of stem cell-derived hepatocytes differentiation project, in collaboration with NIDDK. Purpose and Objectives: The purpose of this acquisition is to purchase the AlphaLISA Albumin Assay kit from PerkinElmer for hepatocyte differentiation project-Screening Compounds for Promoting Hepatocytes Differentiation from Human Stem Cells. We seek the technology/method to test the Albumin secretion level from stem cell-derived hepatocytes in a highly sensitive, quantitative, reproducible and user-friendly mode upon compound library treatment. Project requirements: The hepatocyte differentiation project is a project in collaboration with Dr. Jake Liang's group at NIDDK. Hepatic differentiation of human pluripotent stem cells leads always to generation of hepatocytes like cells (HLCs), which retaining a strong fetal phenotype (maintenance of AFP expression, low albumin (ALB) production). In this context, we hypothesized that small compounds may have an improving effect on the level of cell maturation of the HLCs. In order to perform a high throughput screening of small compounds on stem cell-derived HLCs, our collaborator, Dr. Liang's lab has recently developed an optimized protocol (Carpentier, et al. Stem cells reports, in revision.) allowing the differentiation HLCs in a reproducible and consistent way, suitable for high throughput assay. We plan to treat the HLCs, cultured in 384 well plates, with different compounds from NCATS compound library. In order to assess the effect of the compounds on the level of maturation of the cells, we are planning to measure the secretion of Albumin by using the AlphaLISA technology. It will give us the unique opportunity to assess secreted Albumin without having to transfer supernatants or do wash steps as for ELISA, thus allowing increased consistency. The ultimate purpose of this project is to identify some hits compounds which can significantly promote stem cell-derived hepatocyte differentiation/maturation. The primary hits identified will be then individually validated using a series of phenotypic (percentage of positive cells for hepatic mature markers) and functional (CYP450 metabolism) hepatic assays. Currently, our collaborator at NIDDK has performed preliminary testing to determine the Albumin secretion level of iPS-derived hepatocyte on 384 well plate using the AlphaLISA Albumin kit, which showed high sensitivity and consistency. We will need the same assay format to provide the comparable data for the selectivity. PerkinElmer Inc. has provided us with a significant 40% discount and customizes the AlphaLISA Albumin kits (5000 points/kit) based on our high throughput compound library screening project needs. Anticipated period of performance: Reagent kit will be delivered in a week after receipt of order. The respondent must also provide their DUNS number, organization name, address, point of contact, and size and type of business (e.g., 8(a), HubZone, etc.) pursuant to the applicable NAICS code and any other information that may be helpful in developing or finalizing the acquisition requirements. One (1) copy of the response is required and must be in Microsoft Word or Adobe PDF format using 11-point or 12-point font, 8-1/2" x 11" paper size, with 1" top, bottom, left and right margins, and with single or double spacing. The information submitted must be must be in and outline format that addresses each of the elements of the project requirement and in the capability statement /information sought paragraphs stated herein. A cover page and an executive summary may be included but is not required. The response must include the respondents' technical and administrative points of contact, including names, titles, addresses, telephone and fax numbers, and e-mail addresses. All responses to this notice must be submitted electronically to the Contract Specialist and Contracting Officer. Facsimile responses are NOT accepted. The response must be submitted to Rodney E. Brooks, Contract Specialist,at e-mail address rodney.brooks@nih.gov. The response must be received on or before July 13, 2015, 12 pm, eastern time.
