Description
This is AMENDMENT 2 with revised specifications addressing questions/concerns that were submitted for the services requested. Please acknowledge in your bid package that you are responding to the revised specifications in AMENDMENT 2. Purpose of the requested sequencing service The purpose of the requested sequencing service is to obtain full length sequences of 48 BAC clones with a combination strategy of PacBio and subcloning sequencing. Each BAC clone contains approximately 150 kB insert DNA fragment. The combination of PacBio and subcloning sequencing is required because these BAC clones contain short (about 1 kB), highly repeated regions. PacBio sequencing is to overcome the assembly problem whereas subcloning sequencing is to guarantee sequence quality. Past effort with subcloning sequence alone failed to assemble the subclone sequences. Detailed specifications for the sequencing service project 1) The project includes sequencing 48 BAC clones with insert size ~150 kB in the vector pECBA1. The BAC library will be delivered to the contractor for picking up individual BAC clone for the project. A list of the 48 BAC clones will be provided. 2) The project includes DNA isolation, PacBio sequencing library, PacBio SMRT cell sequencing, and automated assembly and mapping. 3) Make NGS grade BAC DNA. 4) Prepare ONE NGS library of every 12 pooled BAC clones. 5) Prepare individual tagged NGS BAC clone libraries for a run on a MiSeq. 6) Provide one PacBio run and one MiSeq run for each 12 pooled BAC clones. 7) Deliver complete, assembled 48 BAC clone sequences within 15 weeks after receiving the BAC clone samples. 8) Finished data should be delivered via downloadable, password protected FTP server.
